Description
Overview of the medicine
Leaxe 335 gm Oral Solution is a bowel cleansing agent used to empty the colon before a colonoscopy, surgery, or other intestinal procedures. It can also be used for the treatment of chronic constipation. It works by drawing water into the colon, which softens the stool and promotes bowel movements.
Uses & Indications
Dosage
Adults
For bowel cleansing: Dissolve the contents of one 335 gm sachet in 2-4 liters of water and drink according to a specific regimen (e.g., 250 ml every 10-15 minutes until consumed or rectal effluent is clear). For chronic constipation: 1-2 sachets (smaller doses) daily dissolved in water.
Elderly
Same as adult dosage, but monitor for dehydration and electrolyte imbalance, especially in frail patients.
Renal_impairment
Use with caution in severe renal impairment; monitor electrolyte levels. Generally considered safe due to minimal systemic absorption, but high volume intake can be a concern.
How to Take
Dissolve the entire contents of the sachet in the recommended amount of water as directed by the physician or product instructions. Drink the solution over a specified period. For bowel preparation, it is usually taken in divided doses the evening before and the morning of the procedure.
Mechanism of Action
Polyethylene Glycol (PEG) 3350 is an osmotic laxative. It works by retaining water in the intestinal lumen, thereby increasing the water content of the stool. This results in a softer stool consistency and increased stool volume, stimulating peristalsis and facilitating bowel evacuation. The electrolytes help prevent significant fluid and electrolyte shifts.
Pharmacokinetics
Onset
Bowel evacuation usually begins within 1-3 hours after the first dose.
Excretion
Excreted unchanged primarily in the feces.
Half life
Not applicable as systemic absorption is minimal.
Absorption
Minimally absorbed from the gastrointestinal tract (less than 1%).
Metabolism
Not metabolized by gut flora or enzymes.
Side Effects
Contraindications
- Gastrointestinal obstruction or perforation.
- Toxic megacolon or ileus.
- Known hypersensitivity to PEG or any component.
- Severe active inflammatory bowel disease.
Drug Interactions
Oral medications
Leaxe can reduce the absorption of other orally administered drugs due to rapid bowel transit. Administer other oral medications at least 1 hour before starting Leaxe or wait until after the bowel cleansing is complete.
Diuretics, ACE inhibitors, ARBs
Increased risk of electrolyte imbalance, especially hypokalemia, with concomitant use.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children. Once reconstituted, store the solution in a refrigerator and use within 24-48 hours.
Overdose
Symptoms of overdose include severe diarrhea, dehydration, and electrolyte imbalance. Treatment involves supportive care, fluid and electrolyte replacement.
Pregnancy & Lactation
Generally considered safe for use during pregnancy and lactation as systemic absorption is minimal. Consult a doctor.

Lab Monitoring
- Electrolyte levels (Potassium, Sodium) for patients with renal impairment, cardiac disease, or those on diuretics.
- Renal function (Creatinine, BUN) in at-risk patients.
Doctor Notes
- Ensure patients understand the importance of complete consumption of the solution for effective bowel cleansing.
- Monitor high-risk patients (elderly, renal/cardiac impairment) for fluid and electrolyte disturbances.
- Advise patients on dietary restrictions before and during the procedure.
Patient Guidelines
- Follow the instructions precisely for bowel preparation to ensure a successful procedure.
- Drink plenty of clear liquids to prevent dehydration.
- Report any severe abdominal pain, excessive bloating, or allergic reactions to your doctor.
Missed Dose Advice
For bowel preparation, it is crucial to complete the entire course as directed to ensure proper cleansing. If a dose is missed, contact your doctor immediately as the procedure may need to be rescheduled. For chronic constipation, take the missed dose as soon as you remember, but do not double the dose. Continue with your regular schedule.
Driving Precautions
Leaxe generally does not impair the ability to drive or operate machinery. However, due to the urgent need for bowel movements during bowel preparation, driving should be avoided.
Lifestyle Advice
- For chronic constipation, also incorporate dietary fiber, adequate fluid intake, and regular exercise.
- Maintain good hydration throughout the day, especially when using laxatives.
