Overview of the medicine
Letrocare 2.5 mg Tablet contains Letrozole, which is an aromatase inhibitor used to treat breast cancer in postmenopausal women. It works by reducing the amount of estrogen produced by the body, which can help slow or stop the growth of certain types of breast cancer.
Uses & Indications
Dosage
Adults
The recommended dose is 2.5 mg once daily. Treatment should continue for as long as advised by the doctor, typically for 5 years or until tumor recurrence.
Elderly
No dose adjustment is required for elderly patients.
Renal_impairment
No dose adjustment is necessary for patients with mild to moderate renal impairment. Caution is advised in patients with severe renal impairment (creatinine clearance <10 mL/min).
How to Take
Letrocare tablet should be taken orally once daily, with or without food, at approximately the same time each day.
Mechanism of Action
Letrozole is a non-steroidal aromatase inhibitor. It selectively and competitively inhibits the aromatase enzyme, which is responsible for converting androgens into estrogens. By blocking aromatase, Letrozole significantly lowers circulating estrogen levels, thereby removing the estrogenic stimulus for growth of hormone-sensitive breast cancer cells.
Pharmacokinetics
Onset
Estrogen suppression begins within 24-48 hours.
Excretion
Mainly renal excretion (approximately 60% of the dose as the inactive metabolite); about 3.7% is excreted in feces. Less than 0.2% of the dose is excreted unchanged in urine.
Half life
Approximately 2 days (48 hours). Steady-state plasma concentrations are reached after 2-6 weeks.
Absorption
Rapidly and well absorbed from the gastrointestinal tract; absolute bioavailability is approximately 99.9%. Peak plasma concentrations are reached in 1-2 hours.
Metabolism
Primarily hepatic metabolism via cytochrome P450 isoenzymes CYP2A6 and CYP3A4 to an inactive carbinol metabolite.

Contraindications

Drug Interactions
Tamoxifen
Concurrent use with tamoxifen or other antiestrogens should be avoided as it may reduce Letrozole's efficacy.
Estrogen-containing medications
Avoid co-administration with estrogen-containing drugs as they would counteract the pharmacological action of Letrozole.
CYP3A4 and CYP2A6 inhibitors/inducers
Caution with potent inhibitors or inducers of CYP3A4 and CYP2A6 as they may affect Letrozole metabolism.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
There is no specific antidote for Letrozole overdose. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Letrocare is contraindicated in pregnant and lactating women. Women of childbearing potential should use effective contraception during treatment and for at least three weeks after the last dose.
