Description
Overview of the medicine
Levostin 500 mg Tablet contains Levofloxacin, a fluoroquinolone antibiotic used to treat a wide range of bacterial infections. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
Usually 500 mg orally once daily for 7-14 days depending on the infection type and severity.
Elderly
Dosage adjustment may be necessary in elderly patients with impaired renal function.
Renal_impairment
Dosage reduction is required for patients with creatinine clearance less than 50 mL/min. Refer to specific guidelines.
How to Take
Levostin 500 mg Tablet can be taken orally with or without food. Ensure adequate hydration.
Mechanism of Action
Levofloxacin inhibits bacterial DNA gyrase and topoisomerase IV, essential enzymes involved in bacterial DNA replication, transcription, repair, and recombination. This leads to disruption of bacterial DNA and cell death.
Pharmacokinetics
Onset
Within 1-2 hours of administration.
Excretion
Primarily excreted unchanged in the urine, mainly via glomerular filtration and tubular secretion.
Half life
Approximately 6-8 hours.
Absorption
Rapid and nearly complete oral absorption, bioavailability approximately 99%. Peak plasma concentrations achieved within 1-2 hours.
Metabolism
Minimally metabolized in the liver (less than 5% of a dose).
Side Effects
Contraindications
- Hypersensitivity to levofloxacin, other quinolones, or any excipients.
- History of tendinitis or tendon rupture associated with fluoroquinolone use.
- Epilepsy.
- Children and adolescents (under 18 years of age).
Drug Interactions
NSAIDs
Increased risk of CNS stimulation and convulsive seizures.
Warfarin
Enhanced anticoagulant effects, leading to an increased risk of bleeding. Monitor INR closely.
Corticosteroids
Increased risk of severe tendon disorders (especially tendon rupture).
Antacids, Sucralfate, Metal Cations (e.g., iron, zinc)
May significantly decrease levofloxacin absorption if taken concurrently. Administer levofloxacin at least 2 hours before or 2 hours after these agents.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include confusion, dizziness, impaired consciousness, convulsive seizures, and cardiac abnormalities. In case of overdose, gastric lavage and symptomatic and supportive treatment should be initiated. Monitor ECG due to the potential for QT prolongation.
Pregnancy & Lactation
Pregnancy Category C. Levofloxacin should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is excreted in human milk, and due to the potential for serious adverse reactions in nursing infants, a decision should be made whether to discontinue nursing or discontinue the drug, taking into account the importance of the drug to the mother.
