Description
Overview of the medicine
Limbix 125 mg Tablet is an acetylcholinesterase inhibitor primarily used for the symptomatic treatment of mild to moderate Alzheimer's disease. It helps improve cognitive function by increasing the amount of acetylcholine in the brain.
Uses & Indications
Dosage
Adults
Initially 125 mg once daily at bedtime. May be increased to 250 mg daily if tolerated and needed after 4-6 weeks.
Elderly
Same as adult dosage, with careful monitoring for side effects.
Renal_impairment
No dosage adjustment required for mild to moderate renal impairment. Use with caution in severe impairment.
How to Take
Administer orally once daily in the evening, just before bedtime, with or without food. Swallow the tablet whole with water. Do not crush or chew.
Mechanism of Action
Limbix reversibly inhibits the enzyme acetylcholinesterase, which breaks down acetylcholine. By preventing the breakdown of acetylcholine, it increases its concentration in the synaptic cleft, thereby enhancing cholinergic neurotransmission in the central nervous system.
Pharmacokinetics
Onset
Therapeutic effects may be observed within 2-4 weeks, with full effect after several months of consistent dosing.
Excretion
Eliminated predominantly in the urine (57%) and feces (21%) as unchanged drug and metabolites.
Half life
Approximately 70 hours.
Absorption
Rapidly and almost completely absorbed after oral administration, reaching peak plasma concentrations in 3-5 hours.
Metabolism
Primarily metabolized in the liver by cytochrome P450 isoenzymes (CYP2D6 and CYP3A4) to several major metabolites, some of which are active.
Side Effects
Contraindications
- •Hypersensitivity to donepezil, piperidine derivatives, or any component of the formulation
- •Severe gastrointestinal obstruction
Drug Interactions
NSAIDs
Increased risk of gastrointestinal bleeding.
Rifampicin, Phenytoin
May decrease plasma concentrations of Limbix due to induction of CYP enzymes.
Ketoconazole, Quinidine
May increase plasma concentrations of Limbix due to inhibition of CYP3A4 and CYP2D6.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms include severe nausea, vomiting, salivation, sweating, bradycardia, hypotension, seizures, and increased muscle weakness. Treatment should be supportive and involve an anticholinergic agent like atropine sulfate.
Pregnancy & Lactation
Pregnancy Category C. Use only if potential benefit justifies potential risk to the fetus. It is not known whether Limbix is excreted in human milk. Exercise caution when administered to a nursing woman.
