Description
Overview of the medicine
Lipicut 10 mg Tablet contains Atorvastatin, a statin medication used to lower high cholesterol and triglyceride levels in the blood, thereby reducing the risk of heart disease and stroke.
Uses & Indications
Dosage
Adults
Initial dose is usually 10-20 mg once daily. May be increased to a maximum of 80 mg once daily based on LDL-C goals and patient response. Administer at any time of day with or without food.
Elderly
No dosage adjustment required for elderly patients.
Renal_impairment
No dosage adjustment required for renal impairment.
How to Take
Administer orally once daily, at any time of day, with or without food.
Mechanism of Action
Atorvastatin selectively inhibits HMG-CoA reductase, an enzyme crucial for cholesterol synthesis in the liver. This inhibition reduces cholesterol production, leading to increased LDL receptor expression on hepatocytes and enhanced clearance of LDL-cholesterol from the bloodstream.
Pharmacokinetics
Onset
Lipid-lowering effects typically observed within 2 weeks, maximal effect usually within 4 weeks.
Excretion
Primarily via biliary excretion after hepatic metabolism.
Half life
Approximately 14 hours (for parent drug) but active metabolites contribute significantly to activity.
Absorption
Rapidly absorbed, peak plasma concentrations within 1-2 hours. Bioavailability approx 12%.
Metabolism
Primarily hepatic via CYP3A4 to active and inactive metabolites.
Side Effects
Contraindications
- Active liver disease or unexplained persistent elevations of serum transaminases.
- Pregnancy and lactation.
- Hypersensitivity to Atorvastatin or any component of the formulation.
Drug Interactions
Warfarin
Enhanced anticoagulant effect.
Cyclosporine, gemfibrozil, other fibrates, niacin (lipid-lowering doses)
Increased risk of myopathy/rhabdomyolysis.
CYP3A4 inhibitors (e.g., clarithromycin, itraconazole, protease inhibitors)
Increased plasma concentrations of Atorvastatin, increasing risk of side effects.
Storage
Store below 30°C in a dry place, away from light and moisture.
Overdose
There is no specific treatment for atorvastatin overdose. In the event of an overdose, the patient should be treated symptomatically, and supportive measures instituted as required. Due to extensive protein binding, hemodialysis is not expected to significantly enhance atorvastatin clearance.
Pregnancy & Lactation
Atorvastatin is contraindicated during pregnancy and lactation. Women of childbearing potential should use adequate contraception during treatment.
