Locopain-TR 100 mg Capsule contains Tramadol Hydrochloride, an opioid analgesic used for the management of moderate to severe chronic pain. The extended-release formulation allows for once-daily dosing.
Uses & Indications
Dosage
Adults
Initial: 100 mg once daily. May be titrated up to 300 mg once daily based on pain and tolerability. Do not exceed 300 mg/day.
Elderly
For patients > 75 years, adjust dosage cautiously. Maximum 300 mg/day.
Renal_impairment
Creatinine clearance < 30 mL/min, dosage adjustment may be required. Typically, 50-100 mg every 12 hours or avoid extended-release formulation.
How to Take
Swallow the capsule whole; do not crush, chew, or split. Can be taken with or without food.

Tramadol acts as a central analgesic with a dual mechanism: it binds to μ-opioid receptors and inhibits the reuptake of norepinephrine and serotonin, enhancing descending pain inhibitory pathways.

- Patients receiving MAO inhibitors or within 14 days of stopping them
- Severe respiratory depression
- Acute bronchial asthma in unmonitored settings or in absence of resuscitative equipment
Drug Interactions
Carbamazepine
Significantly decreases tramadol plasma levels.
MAO Inhibitors
Increased risk of serotonin syndrome and seizures.
Serotonergic Drugs (SSRIs, SNRIs, TCAs)
Increased risk of serotonin syndrome.
CNS Depressants (e.g., Benzodiazepines, Alcohol)
Increased risk of respiratory depression, sedation, coma, and death.
