Description
Overview of the medicine
Lorazepam is a benzodiazepine used to treat anxiety disorders, insomnia, status epilepticus, and for preoperative sedation.
Uses & Indications
Dosage
Adults
Anxiety: 1-4 mg daily in divided doses. Insomnia: 2-4 mg at bedtime. For severe anxiety or specific indications, higher doses like 5 mg might be used under strict medical supervision.
Elderly
Initially 0.5-1 mg daily, adjusted as needed and tolerated due to increased sensitivity and risk of side effects.
Renal_impairment
No specific dosage adjustment required, but use with caution. Consider lower doses if severe impairment.
How to Take
Administer orally with or without food. For insomnia, take shortly before bedtime. Do not crush or chew sustained-release forms (if applicable).
Mechanism of Action
Lorazepam acts by enhancing the effect of gamma-aminobutyric acid (GABA), a neurotransmitter that inhibits activity in the brain, leading to anxiolytic, sedative, hypnotic, and anticonvulsant effects.

15-60 minutes (oral)
Excretion
Mainly renal excretion as glucuronides.
Half life
10-20 hours
Absorption
Rapid and complete absorption after oral administration.

- Acute narrow-angle glaucoma
- Severe respiratory insufficiency
- Sleep apnea syndrome
- Myasthenia gravis

Increases plasma concentration and prolongs the half-life of lorazepam.
Opioids, alcohol, and other CNS depressants
Increased risk of profound sedation, respiratory depression, coma, and death.
Storage
Store at room temperature (below 30°C), protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include drowsiness, confusion, lethargy, ataxia, dysarthria, hypotension, hypotonia, respiratory depression, and coma. Management involves supportive care, maintaining airway, and in severe cases, administration of flumazenil, a benzodiazepine receptor antagonist.
