Loprazol 20 mg capsule contains Omeprazole, a proton pump inhibitor used to reduce stomach acid production and treat conditions like heartburn, acid reflux, and ulcers.
Uses & Indications
Dosage
Adults
Typically 20-40 mg once daily, depending on the condition. For GERD, 20 mg once daily for 4-8 weeks. For duodenal ulcer, 20 mg once daily for 2-4 weeks. For gastric ulcer, 20 mg once daily for 4-8 weeks. For Zollinger-Ellison, individualized, usually 60 mg once daily.
Elderly
No dose adjustment generally required, but caution with severe hepatic impairment.
Renal_impairment

How to Take
Take orally, preferably in the morning before a meal. Swallow whole with water; do not chew, crush, or open the capsule.
Mechanism of Action
Omeprazole inhibits the H+/K+-ATPase enzyme system (proton pump) in gastric parietal cells, thereby blocking the final step of acid production.
Pharmacokinetics
Onset
Within 1 hour; maximal effect within 2 hours.
Excretion
Mainly excreted in urine (approx. 80%) and feces (approx. 20%) as metabolites.
Half life
0.5-1 hour, but duration of acid suppression is much longer (up to 72 hours).
Absorption
Rapidly absorbed after oral administration, bioavailability increases with repeated dosing.
Metabolism
Extensively metabolized in the liver by the cytochrome P450 system (primarily CYP2C19 and CYP3A4).
Side Effects
Contraindications
- •Hypersensitivity to omeprazole or other substituted benzimidazoles.
- •Concomitant use with nelfinavir.
Drug Interactions
Digoxin
May increase digoxin levels due to increased gastric pH.
Diazepam
May prolong elimination of diazepam due to inhibition of CYP2C19.
Warfarin
May increase INR and bleeding risk due to inhibition of CYP2C19.
Clopidogrel
Omeprazole can reduce the antiplatelet effect of clopidogrel by inhibiting CYP2C19.
Methotrexate
May increase methotrexate levels, leading to toxicity, especially with high doses.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose are generally mild and self-limiting, including nausea, vomiting, dizziness, abdominal pain, diarrhea, and headache. Treatment is symptomatic and supportive.
Pregnancy & Lactation
Use during pregnancy only if clearly needed and the potential benefits outweigh the risks. Excreted in breast milk; caution advised, or consider alternative due to potential effects on the infant.
