Maclo 100 mg Tablet is a non-steroidal anti-inflammatory drug (NSAID) used for the relief of pain and inflammation in conditions like osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis.
Uses & Indications
Dosage
Adults
100 mg twice daily, preferably with or after food.
Elderly
No dosage adjustment is generally required, but caution is advised, and the lowest effective dose should be used.
Renal_impairment
No dosage adjustment is usually necessary for mild renal impairment, but caution is advised. Not recommended in severe renal impairment.
How to Take
Take orally with a glass of water, preferably with or after food to minimize gastrointestinal upset.
Mechanism of Action
Aceclofenac works by inhibiting the synthesis of prostaglandins, which are chemicals in the body that cause pain and inflammation. It primarily inhibits cyclooxygenase (COX) enzymes.

Excretion
Approximately two-thirds of the dose is excreted in the urine and one-third in the faeces.

Extensively metabolized in the liver, primarily via CYP2C9, to 4'-hydroxyaceclofenac and other metabolites.

- Severe renal or hepatic impairment
- Severe heart failure
- Third trimester of pregnancy
- History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs

Increased risk of gastrointestinal bleeding.
Lithium
Increased plasma lithium levels.
Methotrexate
Increased methotrexate toxicity.
Corticosteroids
Increased risk of gastrointestinal ulceration or bleeding.
Diuretics and ACE inhibitors
Reduced hypotensive effect and increased risk of renal impairment.
Warfarin and other anticoagulants
Increased risk of bleeding.

Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.

Symptoms may include headache, nausea, vomiting, epigastric pain, gastrointestinal bleeding. Management is symptomatic and supportive. Gastric lavage and activated charcoal may be considered within one hour of ingestion.
Pregnancy & Lactation
Avoid during the third trimester of pregnancy due to potential fetal harm. Use with caution during the first two trimesters only if the potential benefit outweighs the risk. Not recommended during breastfeeding as aceclofenac may pass into breast milk.
