Description
Overview of the medicine
Mecozol (Omeprazole) is a proton pump inhibitor that reduces the amount of acid produced in the stomach. It is used to treat conditions such as stomach ulcers, gastroesophageal reflux disease (GERD), and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
For GERD: 20-40 mg once daily for 4-8 weeks. For duodenal ulcer: 20 mg once daily for 2-4 weeks. For gastric ulcer: 20 mg once daily for 4-8 weeks. For H. pylori eradication: 20 mg twice daily in combination with antibiotics.
Elderly
No specific dose adjustment is required, but caution is advised in elderly patients with hepatic impairment.
Renal_impairment
No dose adjustment is necessary for patients with impaired renal function.
How to Take
Take orally, usually once daily in the morning, before a meal. Swallow the capsule whole with water; do not chew or crush.
Mechanism of Action
Omeprazole works by irreversibly binding to and inhibiting the H+/K+-ATPase enzyme (the proton pump) in the gastric parietal cells, thereby suppressing gastric acid secretion.
Pharmacokinetics
Onset
Onset of acid suppression occurs within 1 hour, with maximum effect within 2 hours.
Excretion
Approximately 80% of metabolites are excreted in the urine and the remainder in the feces.
Half life
Plasma half-life is short, about 0.5 to 1 hour, but the duration of acid suppression is much longer due to irreversible binding to the proton pump.
Absorption
Rapidly absorbed from the gastrointestinal tract, with peak plasma concentrations occurring within 0.5 to 3.5 hours. Bioavailability is approximately 30-40%.
Metabolism
Extensively metabolized in the liver, primarily by CYP2C19 and CYP3A4 enzymes.
Side Effects
Contraindications
- Hypersensitivity to omeprazole or any other proton pump inhibitors.
- Coadministration with nelfinavir.
Drug Interactions
Diazepam
Omeprazole can prolong the elimination of diazepam.
Warfarin
May increase INR and prothrombin time, requiring monitoring.
Clopidogrel
Concomitant use may reduce the antiplatelet effect of clopidogrel.
Methotrexate
Concomitant use may increase and prolong serum levels of methotrexate.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include nausea, vomiting, dizziness, abdominal pain, diarrhea, and headache. Treatment is symptomatic and supportive.

Pregnancy: Category C. Use only if potential benefit justifies potential risk to the fetus. Lactation: Omeprazole is excreted in human milk. Caution should be exercised when administered to a nursing mother.
