Description
Overview of the medicine
Motilex 10 mg Tablet contains Mosapride Citrate, a prokinetic agent that helps improve gastrointestinal motility. It is primarily used to relieve symptoms associated with chronic gastritis, such as heartburn, nausea, and bloating.
Uses & Indications
Dosage
Adults
The usual adult dose is 5 mg (as Mosapride Citrate) three times a day before or after meals. For 10 mg tablet, dosage should be strictly as prescribed by a physician, typically 10 mg once daily or 5 mg twice daily depending on individual needs and formulation (e.g., extended-release).
Elderly
No specific dose adjustment is generally required, but caution is advised. Dosage should be determined by a physician.
Renal_impairment
Caution is advised. Dosage reduction may be necessary in patients with severe renal impairment. Consult a physician.
How to Take
Take the tablet orally with water. It can be taken before or after meals, as directed by your physician.
Mechanism of Action
Mosapride is a selective 5-HT4 receptor agonist that enhances gastrointestinal motility by increasing acetylcholine release from enteric neurons. This leads to increased gastric emptying and improved coordination of gut movements.
Pharmacokinetics
Onset
Clinical effects may be observed within 30-60 minutes.
Excretion
Mainly excreted via urine (approximately 70% as metabolites) and feces.
Half life
The elimination half-life of mosapride itself is about 1.5-2 hours, while its main active metabolite (M-1) has a half-life of approximately 7 hours.
Absorption
Rapidly absorbed after oral administration, with peak plasma concentrations reached within 1-2 hours.
Metabolism
Extensively metabolized in the liver, primarily via CYP3A4-mediated oxidation and subsequent hydroxylation.
Side Effects
Contraindications
- •Hypersensitivity to mosapride or any component of the tablet.
- •Gastrointestinal hemorrhage, mechanical obstruction, or perforation.
Drug Interactions
Anticholinergic agents
Anticholinergic agents (e.g., atropine, scopolamine) may reduce the prokinetic effect of mosapride.
Azole antifungals/Macrolide antibiotics
Concomitant use with strong CYP3A4 inhibitors (e.g., ketoconazole, erythromycin) may increase mosapride plasma concentrations.
Storage
Store below 30°C, in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptomatic and supportive measures should be taken. There is no specific antidote.
Pregnancy & Lactation
Pregnancy Category C. Use in pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known whether mosapride is excreted in human milk; caution should be exercised when administered to a nursing mother.
