Description
Overview of the medicine
Nasox 500 mg tablet contains Naproxen, a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, inflammation, and fever. It is commonly prescribed for conditions like arthritis, menstrual pain, and muscle aches.
Uses & Indications
Dosage
Adults
For mild to moderate pain, dysmenorrhea, or acute tendinitis/bursitis: 500 mg initially, followed by 250 mg every 6-8 hours as needed. Maximum daily dose: 1250 mg. For chronic conditions like arthritis: 250 mg to 500 mg twice daily.
Elderly
Use the lowest effective dose for the shortest duration. Monitor for adverse effects, especially gastrointestinal and renal.
Renal_impairment
Dosage reduction may be necessary. Not recommended for severe renal impairment (creatinine clearance < 30 mL/min).
How to Take
Take orally with food or milk to minimize gastrointestinal upset. Swallow the tablet whole with a glass of water, do not crush or chew.
Mechanism of Action
Naproxen works by inhibiting the activity of cyclooxygenase (COX-1 and COX-2) enzymes, which are involved in the synthesis of prostaglandins. Prostaglandins are mediators of pain, inflammation, and fever.
Pharmacokinetics
Onset
Pain relief usually within 1 hour; anti-inflammatory effects may take longer, up to 1-2 weeks for full effect in chronic conditions.
Excretion
Primarily excreted in the urine (approximately 95% as naproxen, 6-O-desmethylnaproxen, and their conjugates).
Half life
Approximately 12-17 hours.
Absorption
Rapidly and completely absorbed from the gastrointestinal tract. Peak plasma concentrations occur within 2-4 hours.
Metabolism
Extensively metabolized in the liver to 6-O-desmethylnaproxen, primarily via CYP2C9 and CYP2C18, and subsequently glucuronidated.
