Overview of the medicine
Naspro Plus 375 mg Tablet is a combination medicine containing Naproxen, a non-steroidal anti-inflammatory drug (NSAID), and Esomeprazole, a proton pump inhibitor (PPI). It is used to relieve pain and inflammation while protecting the stomach from the irritating effects of Naproxen.
Uses & Indications
Dosage
Adults
One tablet (Naproxen 375 mg/Esomeprazole 20 mg) twice daily, at least 30 minutes before meals.
Elderly
Use with caution; lower doses may be considered. Refer to physician's advice.
Renal_impairment
Contraindicated in severe renal impairment. Use with caution in mild to moderate impairment with reduced dose.
How to Take
Swallow the tablet whole with water, at least 30 minutes before a meal. Do not chew, crush, or split the tablet.
Mechanism of Action
Naproxen inhibits cyclooxygenase (COX-1 and COX-2) enzymes, leading to reduced prostaglandin synthesis, which mediates pain and inflammation. Esomeprazole is a proton pump inhibitor that reduces gastric acid secretion by irreversibly binding to and inhibiting the H+/K+-ATPase pump in gastric parietal cells.

Onset
Naproxen: 1 hour; Esomeprazole: 1 hour for acid suppression
Excretion
Naproxen and its metabolites are primarily excreted in the urine. Esomeprazole metabolites are primarily excreted in urine and feces.
Half life
Naproxen: 12-17 hours; Esomeprazole: 1-1.5 hours
Absorption
Naproxen is rapidly and completely absorbed from the GI tract. Esomeprazole is also rapidly absorbed.
Metabolism
Both Naproxen and Esomeprazole are primarily metabolized in the liver by cytochrome P450 enzymes.
Side Effects
Contraindications
- •Known hypersensitivity to Naproxen, Esomeprazole, or any component of the formulation
- •History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- •Severe renal impairment
- •Late pregnancy (third trimester)
Drug Interactions
Digoxin
Increased serum digoxin levels.
Warfarin
Increased risk of bleeding.
Clopidogrel
Increased risk of bleeding.
SSRIs/SNRIs
Increased risk of gastrointestinal bleeding.
Methotrexate
Increased methotrexate toxicity.
Diuretics/ACE Inhibitors
Reduced hypotensive effects and increased risk of renal impairment.
Storage
Store below 30°C, in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of Naproxen overdose include lethargy, drowsiness, epigastric pain, nausea, vomiting, and GI bleeding. Esomeprazole overdose symptoms are generally mild and include temporary GI disturbances. Treatment is symptomatic and supportive; gastric lavage and activated charcoal may be considered.
Pregnancy & Lactation
Pregnancy Category C (first and second trimesters), Category D (third trimester for Naproxen - risk of premature closure of ductus arteriosus). Esomeprazole is Pregnancy Category C. Avoid use during late pregnancy. Consult doctor during lactation.
