Overview of the medicine
Nerupa 10 mg Tablet contains Aripiprazole, an atypical antipsychotic medication used to treat certain mental or mood disorders such as schizophrenia, bipolar disorder, and depression. It helps to restore the balance of certain natural substances in the brain (neurotransmitters) to improve mood, thinking, and behavior.
Uses & Indications
Dosage
Adults
Schizophrenia: Initially 10-15 mg once daily, target dose 10-15 mg once daily. Max 30 mg/day. Bipolar I Disorder: Initially 15 mg once daily, target dose 15 mg once daily. Max 30 mg/day. Adjunctive Depression: Initially 2-5 mg once daily, target dose 5-15 mg once daily.
Elderly
No specific dose adjustment is generally recommended based on age alone, but caution is advised. Start with lower doses and titrate gradually.
Renal_impairment
No dosage adjustment is required for patients with renal impairment.
How to Take
Take orally once daily, with or without food. Swallow the tablet whole with water.
Mechanism of Action
Aripiprazole is thought to act as a partial agonist at dopamine D2 and serotonin 5-HT1A receptors, and as an antagonist at serotonin 5-HT2A receptors. This unique mechanism is believed to contribute to its efficacy in stabilizing dopamine and serotonin systems.

Metabolism
Extensively metabolized in the liver, primarily by CYP2D6 and CYP3A4 enzymes. Dehydro-aripiprazole is the only active metabolite.

Contraindications
- •Known hypersensitivity to aripiprazole or any component of the formulation.
- •History of neuroleptic malignant syndrome (NMS).

Storage

Symptoms of overdose include somnolence, vomiting, tremor, and tachycardia. Management involves supportive care, maintaining airway, ventilation, and oxygenation. Activated charcoal may be useful.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit outweighs the potential risk to the fetus. Not recommended during breastfeeding as it is excreted in human milk.
