Description
Overview of the medicine
Nervopex 100 mg Tablet contains Pregabalin, an anticonvulsant and neuropathic pain medication. It is used to treat neuropathic pain, fibromyalgia, and as an adjunct therapy for partial seizures in adults.
Uses & Indications
Dosage
Adults
Initial dose is usually 75 mg twice daily or 50 mg three times daily, adjusted based on response and tolerability, up to a maximum of 300 mg twice daily or 200 mg three times daily (total 600 mg/day).
Elderly
Dosage adjustment may be necessary due to potential age-related decline in renal function. Start with a lower dose.
Renal_impairment
Dosage must be reduced in patients with impaired renal function based on creatinine clearance. For CrCl 30-60 mL/min, maximum dose is 300 mg/day. For CrCl <15 mL/min, maximum dose is 75 mg/day (or equivalent).
How to Take
Can be taken with or without food. Swallow the tablet whole with water. Do not crush, chew, or break.
Mechanism of Action
Pregabalin binds to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system, reducing the release of several neurotransmitters including glutamate, norepinephrine, and substance P. This action contributes to its anticonvulsant and analgesic effects.
Pharmacokinetics
Onset
Pain relief may be experienced within 1 week for some indications, full effect might take longer.
Excretion
Primarily excreted unchanged in the urine. Renal excretion is the major route of elimination.
Half life
Approximately 6.3 hours.
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations are reached within 1.5 hours.
Metabolism
Negligibly metabolized in humans; less than 0.1% of the dose is metabolized.
Side Effects
Contraindications
- •Hypersensitivity to Pregabalin or any component of the formulation
Drug Interactions
Alcohol
Potentiates the sedative effects of alcohol. Avoid concomitant use.
Opioids (e.g., Oxycodone)
Increased risk of central nervous system (CNS) depression, respiratory depression, and sedation. Concomitant use should be with caution and reduced dosages.
Benzodiazepines (e.g., Lorazepam)
Additive CNS depressant effects. Increased risk of somnolence and sedation.
Thiazolidinediones (e.g., Pioglitazone)
May increase risk of weight gain and peripheral edema.
Storage
Store at controlled room temperature (20°C to 25°C), away from moisture and light. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, agitation, and restlessness. Management is primarily supportive; gastric lavage may be considered if recent ingestion. Hemodialysis is effective.
Pregnancy & Lactation
Pregnancy Category C: Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Not recommended during breastfeeding as it is excreted in breast milk.
