Description
Overview of the medicine
Nispore-03 Eye Drop contains Gatifloxacin 0.3%, a fluoroquinolone antibiotic, used to treat bacterial conjunctivitis and other susceptible ocular infections.
Uses & Indications
Dosage
Adults
Days 1-2: Instill one drop into the affected eye(s) every 2 hours while awake, up to 8 times daily. Days 3-7: Instill one drop into the affected eye(s) 4 times daily.
Elderly
No specific dosage adjustment is required for elderly patients.
Renal_impairment
No specific dosage adjustment is necessary for patients with renal impairment due to minimal systemic absorption.
How to Take
For ophthalmic use only. Wash hands before use. Tilt head back, gently pull down the lower eyelid to form a pocket, instill one drop into the pocket, and then close the eye gently. Do not blink excessively. Avoid touching the dropper tip to the eye or any other surface to prevent contamination.
Mechanism of Action
Gatifloxacin inhibits bacterial DNA gyrase and topoisomerase IV, essential enzymes involved in bacterial DNA replication, transcription, repair, and recombination, leading to bacterial cell death.
Pharmacokinetics
Onset
Rapid onset of action in the ocular tissues.
Excretion
Primarily excreted via the kidneys as unchanged drug after systemic absorption.
Half life
Systemic half-life is approximately 7-14 hours, but local ocular kinetics are more relevant for efficacy.
Absorption
Minimal systemic absorption occurs after ocular administration. Peak plasma concentrations are very low and often below the quantifiable limit.
Metabolism
Minimally metabolized; predominantly excreted unchanged.
Side Effects
Contraindications
- Hypersensitivity to gatifloxacin, other fluoroquinolones, or any component of the medication.
Drug Interactions
Systemic Medications
Due to minimal systemic absorption, clinically significant systemic drug interactions are unlikely.
Other Ophthalmic Medications
If using other ophthalmic drops, allow at least 5 minutes between administrations to prevent dilution or wash-out of Nispore-03.
Storage
Store in a cool, dry place, below 30°C. Protect from light. Do not freeze. Keep out of reach of children.
Overdose
Ocular overdose is unlikely to cause systemic toxicity due to minimal absorption. If excessive drops are instilled, rinse the eye with lukewarm water.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Exercise caution when Nispore-03 is administered to a nursing mother, as it is unknown whether gatifloxacin is excreted in human milk.
