Description
Overview of the medicine
Desloratadine is a non-drowsy antihistamine used to relieve allergy symptoms such as sneezing, runny nose, watery eyes, and itching. It is also used to treat chronic idiopathic urticaria (hives).
Uses & Indications
Dosage
Adults
10 mg once daily, with or without food.
Elderly
No dosage adjustment is generally required for elderly patients with normal renal function.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), consider 10 mg every other day.
How to Take
Nohist 10 mg Tablet should be taken orally, swallowed whole with water. It can be taken with or without food.
Mechanism of Action
Desloratadine is a potent, selective, peripheral H1-receptor antagonist. It effectively blocks the action of histamine, a chemical responsible for allergic symptoms, without causing significant drowsiness.
Pharmacokinetics
Onset
Within 1 hour for antihistaminic effects.
Excretion
Eliminated primarily via urine (40%) and feces (40%) as metabolites.
Half life
Approximately 27 hours.
Absorption
Rapidly absorbed after oral administration, reaching peak plasma concentrations in approximately 3 hours.
Metabolism
Extensively metabolized to 3-hydroxydesloratadine, an active metabolite, primarily by cytochrome P450 enzymes.
Side Effects
Contraindications
- Hypersensitivity to Desloratadine, Loratadine, or any component of the formulation.
- Patients with a history of severe allergic reactions to similar medications.
Drug Interactions
Alcohol
No potentiation of alcohol-induced impairment of psychomotor performance was found in clinical trials.
Erythromycin
Co-administration with erythromycin led to increased plasma concentrations of desloratadine and 3-hydroxydesloratadine.
Ketoconazole
Concomitant administration with ketoconazole resulted in increased plasma concentrations of desloratadine and 3-hydroxydesloratadine.
Fluoxetine, Cimetidine
May lead to increased exposure to desloratadine; however, no clinically relevant adverse events were observed.

Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.

Symptoms of overdose may include drowsiness, tachycardia, and headache. In the event of overdose, symptomatic and supportive treatment is recommended. Desloratadine is not removed by hemodialysis.

Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Desloratadine is excreted into breast milk; therefore, a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
