Overview of the medicine
Norglic 80 mg tablet is an oral antidiabetic medication used to lower blood glucose levels in patients with type 2 diabetes mellitus. It contains Gliclazide, a sulfonylurea, which stimulates insulin secretion from the pancreatic beta cells.
Uses & Indications
Dosage
Adults
Initial dose is usually 40-80 mg daily, taken with breakfast. Dosage can be gradually increased in steps of 40-80 mg, up to a maximum of 320 mg daily, usually divided into two doses.
Elderly
Similar to adult dosage, but titration should be done carefully, starting with a lower dose.
Renal_impairment
In mild to moderate renal impairment, no dose adjustment is usually required. In severe impairment, use with caution, and dosage may need to be reduced.
How to Take
Norglic 80 mg tablet should be taken orally, preferably with breakfast. It should be swallowed whole with water and not crushed or chewed.
Mechanism of Action
Gliclazide acts primarily by stimulating insulin release from the pancreatic β-cells. It binds to the sulfonylurea receptor (SUR1) on the β-cell membrane, leading to the closure of ATP-sensitive potassium channels. This depolarization opens voltage-dependent calcium channels, increasing intracellular calcium, which triggers insulin secretion.

Side Effects
Contraindications
- Hypersensitivity to Gliclazide or other sulfonylureas or any excipients.
- Type 1 diabetes mellitus.
- Diabetic ketoacidosis, diabetic precoma and coma.
- Severe renal or hepatic insufficiency.
- Concomitant treatment with miconazole.

Miconazole
Enhances the hypoglycemic effect, potentially leading to severe hypoglycemia.
Beta-blockers
May mask symptoms of hypoglycemia and potentiate the hypoglycemic effect.
Phenylbutazone, Alcohol, Sulfonamides
Potentiate the hypoglycemic effect of Gliclazide.
Corticosteroids, Danazol, Chlorpromazine
May reduce the hypoglycemic effect, requiring higher doses of Gliclazide.
