Description
Overview of the medicine
Desloratadine is a non-drowsy antihistamine used to relieve allergy symptoms such as sneezing, runny nose, itchy or watery eyes, and hives. It is a long-acting selective peripheral H1-receptor antagonist.
Uses & Indications
Dosage
Adults
Usually 5 mg orally once daily. For severe cases or specific conditions, 10 mg once daily may be prescribed by a physician.
Elderly
No specific dose adjustment is usually required for elderly patients with normal renal function. Use with caution in those with renal or hepatic impairment.
Renal_impairment
Dosage adjustment may be necessary. For severe renal impairment, consider 5 mg every other day.
How to Take
Take orally with or without food. Do not crush or chew the tablet; swallow whole.
Mechanism of Action
Desloratadine is a long-acting tricyclic antihistamine with selective peripheral H1-receptor antagonist activity. It inhibits histamine release from mast cells and basophils, and also inhibits a variety of pro-inflammatory mediators.

Onset
Within 30 minutes to 1 hour.
Excretion
Primarily excreted in urine and feces as metabolites.
Half life
Approximately 27 hours.
Absorption
Rapidly absorbed after oral administration. Food does not affect absorption.
Metabolism
Extensively metabolized to 3-hydroxydesloratadine, an active metabolite, via cytochrome P450 enzymes.
Side Effects
Contraindications
- •Hypersensitivity to desloratadine, loratadine, or any component of the formulation.
Drug Interactions
Cimetidine
Increased plasma concentration of desloratadine.
Fluoxetine
Increased plasma concentration of desloratadine.
Erythromycin
Increased plasma concentration of desloratadine.
Ketoconazole
Increased plasma concentration of desloratadine.
Storage
Store below 30°C in a dry place, away from light. Keep out of reach of children.
Overdose
Symptoms may include drowsiness, somnolence, headache, tachycardia. Management is symptomatic and supportive. Gastric lavage and activated charcoal may be considered.
Pregnancy & Lactation
Pregnancy Category C. Use only if potential benefit justifies potential risk to the fetus. Desloratadine is excreted into breast milk; use with caution during lactation.
