Overview of the medicine
Naproxen is a Non-Steroidal Anti-Inflammatory Drug (NSAID) used to relieve pain, inflammation, and fever.
Uses & Indications
Dosage
Adults
Typically 250-500 mg twice daily, or 375 mg twice daily for sustained release formulations, not exceeding 1250 mg daily.
Elderly
Lower doses may be required; monitor for adverse effects.
Renal_impairment
Reduced dosage or avoidance in severe renal impairment; monitor renal function closely.
How to Take
Oral. Should be taken with food or milk to minimize gastrointestinal upset.
Mechanism of Action
Inhibits prostaglandin synthesis by blocking cyclooxygenase (COX-1 and COX-2) enzymes.

Onset
Pain relief within 1 hour.
Excretion
Primarily excreted in the urine (about 95% as metabolites).

12-17 hours.
Absorption
Rapidly and completely absorbed from the gastrointestinal (GI) tract. Peak plasma levels usually within 2-4 hours.
Metabolism
Primarily metabolized in the liver by demethylation and glucuronidation.

- Hypersensitivity to naproxen or other NSAIDs
- Asthma, urticaria, or allergic-type reactions after aspirin or other NSAIDs
- Active gastrointestinal bleeding or ulceration
- Severe heart failure
- Severe renal or hepatic impairment
- Third trimester of pregnancy

Storage

Symptoms may include drowsiness, heartburn, nausea, vomiting, epigastric pain, and GI bleeding. Management includes gastric lavage, activated charcoal, and supportive care.
Pregnancy & Lactation
Avoid in the third trimester of pregnancy due to risk of premature closure of ductus arteriosus. Use with caution during first and second trimesters only if clearly needed. Excreted in breast milk, use with caution or consider alternatives.
