Description
Overview of the medicine
Olacent is an antidepressant medication used to treat major depressive disorder (MDD), generalized anxiety disorder (GAD), and panic disorder. It works by affecting certain chemicals in the brain.
Uses & Indications
Dosage
Adults
Initial dose is 50 mg once daily, gradually increased to 150 mg once daily as needed, based on clinical response and tolerability. Max dose 200 mg/day.
Elderly
Similar to adults, but initiate at a lower dose (e.g., 25 mg once daily) and titrate slowly to minimize adverse effects.
Renal_impairment
Dose adjustment may be necessary for patients with moderate to severe renal impairment. Consult physician for specific recommendations.
How to Take
Take Olacent tablet orally once daily, with or without food. Swallow the tablet whole; do not crush, chew, or divide.
Mechanism of Action
Olacent selectively inhibits the reuptake of serotonin and norepinephrine in the brain, thereby increasing the concentration of these neurotransmitters in the synaptic cleft. This leads to enhanced monoaminergic neurotransmission.

Onset
Therapeutic effects may begin within 1-2 weeks, but full antidepressant effect may take 2-4 weeks or longer.
Excretion
Primarily excreted through urine as metabolites, with a small portion eliminated in feces.
Half life
Approximately 12-15 hours, allowing for once-daily dosing.
Absorption
Rapidly and almost completely absorbed from the gastrointestinal tract. Peak plasma concentrations are reached within 2-4 hours.
Metabolism
Extensively metabolized in the liver, primarily via CYP2D6 and CYP3A4 enzymes, into inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to Olacetam or any component of the formulation.
- Concomitant use with Monoamine Oxidase Inhibitors (MAOIs) or within 14 days of discontinuing MAOI therapy.
- Severe hepatic impairment.
Drug Interactions
MAOIs
Concurrent use is contraindicated due to risk of serotonin syndrome (e.g., hyperthermia, rigidity, myoclonus, autonomic instability).
Warfarin
May increase anticoagulant effect and risk of bleeding; monitor INR closely.
SSRIs/SNRIs
Increased risk of serotonin syndrome; monitor closely or avoid concomitant use.
CNS Depressants (e.g., alcohol, benzodiazepines)
May potentiate sedative effects; use with caution.
Storage
Store in a cool, dry place below 25°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, nausea, vomiting, dizziness, tachycardia, tremor, and possibly seizures or serotonin syndrome. Management is symptomatic and supportive. Maintain an open airway, monitor cardiac and vital signs. Activated charcoal may be considered.
Pregnancy & Lactation
Category C in pregnancy. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown whether Olacent is excreted in human milk; use with caution in nursing mothers. Consult a physician.
