Onvicon 500 mg Suspension contains Amoxicillin, a penicillin-type antibiotic used to treat a variety of bacterial infections.
Uses & Indications
Dosage
Adults
250-500 mg every 8 hours, or 500-875 mg every 12 hours, depending on the severity and type of infection. For severe infections, up to 1 gram every 8 hours.
Elderly
No specific dosage adjustment needed for elderly patients with normal renal function. Dosage adjustment may be required in cases of significant renal impairment.
Renal_impairment
For creatinine clearance 10-30 mL/min, 250-500 mg every 12 hours. For creatinine clearance <10 mL/min, 250-500 mg every 24 hours. Hemodialysis patients: 250-500 mg every 24 hours, with an additional dose during and at the end of dialysis.
How to Take
Shake the bottle well before each use. Measure the prescribed dose using the provided measuring spoon or cup. Can be taken with or without food. Complete the full course of treatment as prescribed by the doctor, even if symptoms improve.
Mechanism of Action
Amoxicillin works by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins (PBPs) located on the inner bacterial cell membrane, which prevents the cross-linking of peptidoglycan polymers, essential for bacterial cell wall strength and integrity. This leads to weakened bacterial cell walls and eventual cell lysis and death.

Excretion
Primarily excreted unchanged by the kidneys via glomerular filtration and tubular secretion. Approximately 60-70% of a dose is excreted in the urine within 6 hours.
Half life
Approximately 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, with bioavailability of approximately 70-90%. Peak plasma concentrations are reached within 1-2 hours after oral administration.
Metabolism
Minimal hepatic metabolism; about 10-25% is metabolized to inactive penicilloic acid.

Contraindications
- Known hypersensitivity to penicillin antibiotics or to any component of the formulation.
- History of severe allergic reaction (e.g., anaphylaxis) to any beta-lactam antibiotic (e.g., cephalosporins, carbapenems).
Drug Interactions
Probenecid
Increases and prolongs plasma levels of amoxicillin by competing for renal tubular secretion.
Allopurinol
Concomitant use with amoxicillin may increase the incidence of skin rash.
Methotrexate
Amoxicillin may reduce the renal excretion of methotrexate, increasing its toxicity.
Warfarin and other oral anticoagulants
May increase the anticoagulant effect, requiring closer monitoring of INR.
Storage
Store dry powder below 30°C, protected from light and moisture. Once reconstituted, store the suspension in a refrigerator (2-8°C) and discard any unused portion after 7 to 14 days, as per specific product instructions.

Symptoms of overdose may include gastrointestinal disturbances such as nausea, vomiting, and diarrhea, and fluid and electrolyte imbalance. In rare cases, renal failure and convulsions may occur. Treatment is symptomatic and supportive. Hemodialysis can remove amoxicillin from circulation.
