Description
Overview of the medicine
Desloratadine is a non-drowsy antihistamine used to relieve allergy symptoms such as sneezing, runny nose, itching, and hives. It works by blocking a natural substance (histamine) that your body makes during an allergic reaction.
Uses & Indications
Dosage
Adults
5 mg (one tablet) once daily.
Elderly
No specific dose adjustment generally required, but caution is advised in elderly patients with renal or hepatic impairment.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), a starting dose of 5 mg every other day is recommended.
How to Take
Orinex 5 mg tablet can be taken orally, with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Desloratadine is a long-acting tricyclic antihistamine with selective peripheral H1-receptor antagonist activity. It inhibits the release of histamine from mast cells and basophils, and also reduces the release of other inflammatory mediators from mast cells.

Excreted equally in urine and feces as metabolites, with a small amount as unchanged drug.

Approximately 27 hours
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations are reached in approximately 3 hours.
Metabolism
Extensively metabolized to 3-hydroxydesloratadine, an active metabolite, primarily by CYP2C8 and CYP3A4.
Side Effects
Contraindications
- •Hypersensitivity to desloratadine, loratadine, or any component of the formulation.
- •Not recommended for children under 6 months of age.
Drug Interactions
Cimetidine
May increase plasma concentrations of desloratadine.
Fluoxetine
May increase plasma concentrations of desloratadine.
Azithromycin
May increase plasma concentrations of desloratadine.
Erythromycin
May increase plasma concentrations of desloratadine.
Ketoconazole
May increase plasma concentrations of desloratadine.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, tachycardia, and headache. In the event of overdose, symptomatic and supportive treatment should be initiated. Desloratadine is not eliminated by hemodialysis.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Desloratadine is excreted into breast milk, so a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
