Orioplex 5 mg tablet is an antidepressant medication used to treat major depressive disorder, generalized anxiety disorder, and panic disorder. It helps restore the balance of a certain natural substance (serotonin) in the brain.
Uses & Indications
Dosage
Adults
Initial dose 5 mg once daily, preferably in the morning. May be increased to 10-20 mg daily after several weeks based on response.
Elderly
Initial dose 5 mg once daily. Max recommended dose is 10 mg daily due to reduced metabolic clearance.
Renal_impairment
No dosage adjustment needed for mild to moderate renal impairment. Use with caution in severe impairment.
How to Take
Take Orioplex 5 mg tablet orally, with or without food. It can be taken at any time of day, but consistency (e.g., always in the morning) is recommended.
Mechanism of Action
Orioplex selectively inhibits the reuptake of serotonin (5-HT) by neurons in the central nervous system, thereby increasing the concentration of 5-HT in the synaptic cleft and enhancing serotonergic neurotransmission.

Initial therapeutic effects may be observed within 1-2 weeks, with full antidepressant effects usually seen after 4-6 weeks of consistent treatment.
Excretion
Mainly excreted in the urine (approximately 60%) and feces (approximately 40%) as metabolites.

Contraindications
- Hypersensitivity to Orioplex or any component of the tablet.
- Concomitant use with Monoamine Oxidase Inhibitors (MAOIs) or within 14 days of discontinuing MAOI therapy.
Drug Interactions
MAOIs
Concurrent use can lead to serious, sometimes fatal, reactions including Serotonin Syndrome.
NSAIDs
Increased risk of gastrointestinal bleeding.
Warfarin
May increase anticoagulant effect, requiring close INR monitoring.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.

Symptoms of overdose may include drowsiness, nausea, vomiting, dizziness, tachycardia, tremor, and rarely, seizures. Treatment is symptomatic and supportive. Maintain an open airway, monitor cardiac and vital signs. Activated charcoal may be considered.
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Not recommended during breastfeeding as it is excreted in breast milk.
