Description
Overview of the medicine
Oxipin 300 mg Tablet is a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, swelling, and stiffness caused by conditions such as osteoarthritis, rheumatoid arthritis, and other musculoskeletal disorders.
Uses & Indications
Dosage
Adults
300 mg once daily, or 150 mg twice daily. Maximum daily dose is 600 mg.
Elderly
Start with lower doses (e.g., 150 mg once daily) due to increased risk of adverse effects.
Renal_impairment
Use with caution; dose reduction may be necessary in severe impairment. Not recommended for severe renal disease.
How to Take
Take orally, preferably with food or milk to minimize gastrointestinal upset. Swallow the tablet whole with a glass of water.
Mechanism of Action
Oxaprozin works by inhibiting the activity of cyclooxygenase (COX) enzymes, particularly COX-2, which are involved in the synthesis of prostaglandins. Prostaglandins are chemicals that contribute to pain, inflammation, and fever.
Pharmacokinetics
Onset
Pain relief usually begins within 1-2 hours.
Excretion
Mainly excreted in the urine (about 60%) and feces (about 35%) as metabolites.
Half life
Approximately 40-60 hours, allowing for once-daily dosing.
Absorption
Well absorbed from the gastrointestinal tract; peak plasma concentrations reached within 3-5 hours.
Metabolism
Extensively metabolized in the liver, primarily via oxidation and glucuronidation.
Side Effects
Contraindications
- Known hypersensitivity to oxaprozin or other NSAIDs
- History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- Active gastrointestinal bleeding or peptic ulcer
- Severe renal or hepatic impairment
- Third trimester of pregnancy
- Coronary artery bypass graft (CABG) surgery
Drug Interactions
SSRIs
Increased risk of gastrointestinal bleeding.
Lithium
Increased plasma lithium levels, leading to toxicity.
Diuretics
May reduce diuretic effect and increase risk of renal impairment.
Methotrexate
Increased methotrexate toxicity.
ACE Inhibitors / ARBs
May reduce antihypertensive effect and increase risk of renal impairment.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, nausea, vomiting, epigastric pain, lethargy, and gastrointestinal bleeding. In severe cases, acute renal failure, respiratory depression, and coma may occur. Management involves supportive care, gastric lavage, and administration of activated charcoal. Hemodialysis is unlikely to be beneficial.
Pregnancy & Lactation
Oxaprozin should be avoided during pregnancy, especially during the third trimester, due to potential risks to the fetus (e.g., premature closure of the ductus arteriosus). It is also not recommended during breastfeeding as it may pass into breast milk.
