Description
Overview of the medicine
Pivasta 2 mg tablet contains Pitavastatin, a statin used to lower high cholesterol levels and reduce the risk of cardiovascular diseases. It works by reducing the production of cholesterol in the liver.
Uses & Indications
Dosage
Adults
Initial dose 1 mg once daily, usually adjusted to 2-4 mg once daily based on LDL-C levels. Maximum 4 mg once daily.
Elderly
No specific dose adjustment required, but caution is advised.
Renal_impairment
Initial dose 1 mg once daily for moderate to severe impairment (CrCl < 60 mL/min). Maximum 2 mg once daily.
How to Take
Take orally once daily, with or without food, preferably at the same time each day. Can be taken in the morning or evening.
Mechanism of Action
Pitavastatin competitively inhibits HMG-CoA reductase, an enzyme that catalyzes the early, rate-limiting step in cholesterol biosynthesis. This leads to a compensatory increase in LDL receptors on hepatocytes, increasing LDL catabolism.
Pharmacokinetics
Onset
Lipid-lowering effects typically observed within 2 weeks, maximal effect usually within 4 weeks.
Excretion
Primarily excreted via bile (feces) as unchanged drug and metabolites. Renal excretion is minor (approximately 15%).
Half life
Approximately 12 hours (terminal half-life).
Absorption
Rapidly absorbed from the gastrointestinal tract; absolute bioavailability is 51%. Peak plasma concentrations achieved within 1 hour.
Metabolism
Primarily metabolized by CYP2C8 and to a lesser extent CYP2C9; not significantly metabolized by CYP3A4.
