Description
Overview of the medicine
Pollival 0.05% Eye Drop contains Azelastine Hydrochloride, an antihistamine and mast cell stabilizer used to relieve ocular symptoms of allergic conjunctivitis such as itching, redness, and watery eyes.
Uses & Indications
Dosage
Adults
Instill 1 drop into each affected eye twice daily (approximately every 8-12 hours).
Elderly
No specific dosage adjustment is required for elderly patients.
Renal_impairment
No dosage adjustment is needed in patients with renal impairment due to minimal systemic absorption.
How to Take
For ophthalmic use only. Tilt head back, pull down the lower eyelid to form a pocket, instill one drop into the pocket, and then close the eye gently. Do not touch the dropper tip to any surface to avoid contamination.
Mechanism of Action
Azelastine acts as a potent, long-acting H1-receptor antagonist and also inhibits the release of histamine and other mediators from mast cells, thereby reducing the allergic response in the eye.
Pharmacokinetics
Onset
Onset of action is rapid, typically within 3 minutes for symptom relief.
Excretion
Excreted primarily in feces, with a smaller portion excreted in urine.
Half life
The systemic elimination half-life of azelastine is approximately 22 hours, but local ocular effects are immediate.
Absorption
Minimal systemic absorption after ophthalmic administration. Peak plasma concentrations are generally low and often below the quantification limit.
Metabolism
Primarily metabolized in the liver by cytochrome P450 enzymes (e.g., CYP1A2, CYP2C19, CYP3A4) to its primary metabolite, desmethylazelastine.
Side Effects
Contraindications
- Hypersensitivity to azelastine hydrochloride or any component of the formulation.
Drug Interactions
Systemic Medications
Due to minimal systemic absorption, drug-drug interactions with systemically administered medications are unlikely. However, caution should be exercised if co-administering with other CNS depressants due to potential additive effects if significant systemic absorption occurs (rare with ophthalmic use).
Storage
Store below 25°C (77°F). Do not freeze. Protect from light. Keep out of reach of children.
Overdose
Overdose is unlikely with ophthalmic use. If accidentally swallowed, symptomatic and supportive treatment should be initiated. No specific antidote is known.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known whether azelastine is excreted in human breast milk; caution should be exercised when administered to a nursing woman.
