Pristil

Take Pristil tablet orally with or without food, usually once daily. Swallow the tablet whole with water.

Pristil (hypothetically Amlodipine) is a dihydropyridine calcium antagonist (calcium ion antagonist or slow-channel blocker) that inhibits the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle. The contractile process of cardiac muscle and vascular smooth muscle is dependent upon the movement of extracellular calcium ions into these cells through specific ion channels. Pristil inhibits calcium ion influx across the cell membrane selectively, with a greater effect on vascular smooth muscle cells than on cardiac muscle cells.

• Hypersensitivity to Pristil or any dihydropyridine derivatives
• Severe hypotension
• Cardiogenic shock
• Obstruction of the outflow tract of the left ventricle (e.g., high grade aortic stenosis)
• Hemodynamically unstable heart failure after acute myocardial infarction

Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.

Overdose may lead to excessive peripheral vasodilation with marked hypotension and possibly reflex tachycardia. Management involves symptomatic and supportive treatment, including monitoring cardiac and respiratory function, elevation of extremities, and administration of IV fluids and vasopressors if needed.

Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known if Pristil is excreted in human milk; caution should be exercised when administered to a nursing woman.
