Description
Overview of the medicine
Progency-10 mg Tablet contains Progenamide Hydrochloride, a novel antihypertensive agent primarily used to manage essential hypertension. It acts by selectively dilating peripheral blood vessels, thereby reducing blood pressure.
Uses & Indications
Dosage
Adults
Initially 10 mg once daily, adjusted based on response up to 20 mg daily.
Elderly
Start with 5 mg once daily, titrate carefully based on individual response and tolerability.
Renal_impairment
No dose adjustment needed for mild to moderate impairment. Use with caution in severe impairment; monitor blood pressure closely.
How to Take
Take orally, preferably at the same time each day, with or without food. Swallow the tablet whole with a glass of water; do not crush or chew.
Mechanism of Action
Progenamide Hydrochloride is a selective peripheral vasodilator that directly relaxes vascular smooth muscle, leading to vasodilation and a reduction in total peripheral resistance. This action helps to lower systemic blood pressure.
Pharmacokinetics
Onset
Within 30-60 minutes.
Excretion
Primarily excreted via urine (60% as metabolites, 20% unchanged drug); remainder in feces.
Half life
Approximately 6-8 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract, bioavailability approximately 80%. Peak plasma concentrations are reached within 1-2 hours.
Metabolism
Extensively metabolized in the liver via CYP3A4 pathway to inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to Progenamide Hydrochloride or any component of the formulation
- Severe symptomatic hypotension
- Cardiogenic shock
- Severe aortic stenosis
Drug Interactions
NSAIDs
May reduce the antihypertensive effect of Progenamide Hydrochloride.
Diuretics
Concomitant use may potentiate hypotensive effects, requiring dose adjustment of either drug.
Beta-blockers
Additive hypotensive effects possible; monitor blood pressure closely.
Storage
Store below 30°C in a dry place. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include severe hypotension, dizziness, profound weakness, and reflex tachycardia. Treatment is supportive and symptomatic. Gastric lavage and activated charcoal may be considered if ingestion is recent. Intravenous fluids and vasopressors may be administered to manage hypotension.
Pregnancy & Lactation
Not recommended during pregnancy unless essential, due to potential fetal risks. Progenamide Hydrochloride may be excreted in breast milk; use with caution in lactating mothers and only after consulting a doctor.
