Description
Overview of the medicine
Prostacin 0.4 mg capsule is an alpha-1 adrenergic blocker used to treat the signs and symptoms of benign prostatic hyperplasia (BPH). It relaxes the muscles in the prostate and bladder neck, making it easier to urinate.
Uses & Indications
Dosage
Adults
0.4 mg once daily, taken approximately 30 minutes after the same meal each day. May be increased to 0.8 mg once daily after 2-4 weeks if needed.
Elderly
No dosage adjustment is required for elderly patients.
Renal_impairment
No dosage adjustment is required for patients with mild to moderate renal impairment. Use with caution in severe renal impairment.
How to Take
Orally, once daily, approximately 30 minutes after the same meal each day. Swallow the capsule whole; do not crush, chew, or open it.
Mechanism of Action
Tamsulosin selectively blocks alpha-1A and alpha-1B adrenergic receptors in the prostate, bladder neck, and prostatic urethra. This blockade causes relaxation of the smooth muscle in these tissues, leading to improved urine flow and reduced BPH symptoms.
Pharmacokinetics
Onset
Symptomatic improvement may be observed within 2-4 weeks, though effects can be seen earlier.
Excretion
Mainly excreted in urine (about 76% as metabolites), with a small amount in feces.
Half life
Approximately 9-15 hours (terminal half-life).
Absorption
Well absorbed from the gastrointestinal tract, nearly 90% bioavailable when taken on an empty stomach. Food slightly reduces and delays absorption.
Metabolism
Extensively metabolized in the liver, primarily by CYP3A4 and CYP2D6.
Side Effects
Contraindications
- Hypersensitivity to Tamsulosin or any component of the formulation
- History of orthostatic hypotension
Drug Interactions
Other alpha-1 blockers

Strong CYP2D6 inhibitors (e.g., paroxetine)
May increase Tamsulosin exposure. Use with caution.
PDE5 inhibitors (e.g., sildenafil, tadalafil)
Increased risk of hypotension. Use with caution, initiate PDE5 inhibitor at lowest dose.
Strong CYP3A4 inhibitors (e.g., ketoconazole)
May increase Tamsulosin exposure. Avoid concomitant use or reduce Tamsulosin dose.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
