Overview of the medicine
Prucon 2 mg tablet contains Prucalopride, a selective serotonin 5-HT4 receptor agonist used for the symptomatic treatment of chronic idiopathic constipation in adults for whom laxatives have failed to provide adequate relief.
Uses & Indications
Dosage
Adults
The recommended dose is 2 mg once daily, taken with or without food.
Elderly
For elderly patients, initiate treatment with 1 mg once daily. If required and well tolerated, the dose can be increased to 2 mg once daily.
Renal_impairment
For patients with severe renal impairment (creatinine clearance <30 mL/min), the recommended dose is 1 mg once daily.
How to Take
Take Prucon 2 mg tablet orally with or without food. Swallow the tablet whole with water; do not crush, chew, or break it.
Mechanism of Action
Prucalopride acts as a highly selective serotonin 5-HT4 receptor agonist. By stimulating these receptors in the gastrointestinal tract, it increases colonic motility and improves bowel transit, leading to increased stool frequency and improved stool consistency.

Clinical effect typically observed within 1 week of treatment initiation.
Excretion
Primarily excreted unchanged (approximately 60% of the dose) in the urine, with a smaller amount excreted in the feces.

The plasma elimination half-life is approximately 24-30 hours.
Absorption
Rapidly absorbed after oral administration. Peak plasma concentration (Cmax) is reached in approximately 2-3 hours.
Metabolism
Minimally metabolized; primarily by CYP enzymes, but significant metabolism is not observed. Major metabolic products are inactive.

- Hypersensitivity to the active substance or to any of the excipients.
- Intestinal perforation or obstruction.
- Severe inflammatory conditions of the intestinal tract such as Crohn's disease, ulcerative colitis, and toxic megacolon/megarectum.
Drug Interactions
Cyclosporine
Concomitant use with cyclosporine (a P-glycoprotein inhibitor) may increase prucalopride exposure; caution advised.
Ketoconazole
Concomitant use with ketoconazole (a potent CYP3A4 inhibitor) may increase prucalopride exposure; caution advised.

Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
