Description
Overview of the medicine
Raboz 20 mg tablet contains Rabeprazole Sodium, a proton pump inhibitor (PPI) that reduces the amount of acid produced in the stomach. It is used to treat acid-related diseases such as GERD, peptic ulcers, and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
GERD: 20 mg once daily for 4-8 weeks. Duodenal ulcer: 20 mg once daily for 4 weeks. Gastric ulcer: 20 mg once daily for 6 weeks. H. pylori: 20 mg twice daily (with antibiotics).
Elderly
No specific dose adjustment generally needed, but monitor carefully for side effects.
Renal_impairment
No dose adjustment needed for patients with renal impairment.
How to Take
Take orally, preferably in the morning before breakfast. Swallow the tablet whole; do not chew, crush, or break.
Mechanism of Action
Rabeprazole works by irreversibly binding to and inhibiting the H+/K+-ATPase enzyme (the proton pump) in the gastric parietal cells. This action blocks the final step of gastric acid secretion, significantly reducing stomach acid levels.
Pharmacokinetics
Onset
Within 1 hour
Excretion
Primarily via urine (approx. 90%), remainder in feces.
Half life
1-2 hours (though duration of action is much longer due to irreversible binding)
Absorption
Rapidly absorbed, peak plasma concentrations in 2-4 hours. Bioavailability approx. 52%.
Metabolism
Extensively metabolized in the liver via CYP2C19 and CYP3A4.
Side Effects
Contraindications
- •Known hypersensitivity to rabeprazole, other benzimidazole derivatives (e.g., omeprazole, lansoprazole), or any component of the formulation.
- •Severe hepatic impairment (use with caution).
Drug Interactions
Digoxin
Increased digoxin levels due to increased gastric pH.
Warfarin
Increased INR/prothrombin time; monitor closely.
Methotrexate
Increased methotrexate levels; monitor for toxicity.
Ketoconazole/Itraconazole
Decreased absorption of these drugs due to increased gastric pH.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose are generally mild and non-specific. Treatment should be symptomatic and supportive. Rabeprazole is not readily dialyzable.
Pregnancy & Lactation
Pregnancy: Category C. Use only if potential benefit outweighs potential risk to the fetus. Lactation: Not known if rabeprazole is excreted in human milk. Exercise caution when administered to a nursing mother.
