Uses & Indications
Dosage
Adults
One tablet daily, preferably with a meal, or as directed by a physician. Treatment course typically lasts 6-12 weeks.
Elderly
No specific dose adjustment required for elderly patients, follow adult dosage.

How to Take

Mechanism of Action
Glucosamine Sulfate stimulates the production of proteoglycans and collagen, essential components of cartilage matrix, thereby helping to rebuild and repair damaged cartilage. Onset

Excretion
Glucosamine: Primarily via urine and feces. Chondroitin: Primarily via urine. Vitamin D3: Metabolites excreted via bile and feces, small amount in urine.
Half life
Glucosamine: 15-17 hours. Chondroitin: Approximately 3-10 hours. Vitamin D3: ~24 hours (initial), metabolites have longer half-lives.
Absorption
Glucosamine: Approximately 90% is absorbed orally. Chondroitin: Low oral bioavailability, around 10-20%. Vitamin D3: Well absorbed orally, fat-soluble.
Metabolism
Glucosamine: Metabolized primarily in the liver, with some metabolism in the intestinal wall. Chondroitin: Partially depolymerized in the gut by bacteria. Vitamin D3: Metabolized in the liver to 25-hydroxyvitamin D [25(OH)D] and then in the kidneys to 1,25-dihydroxyvitamin D [1,25(OH)2D], the active form.
Side Effects
Contraindications

Drug Interactions
Thiazide diuretics
May increase the risk of hypercalcemia with Vitamin D3.
Tetracycline antibiotics
Glucosamine may decrease the absorption of tetracycline. Separate administration by several hours.
Warfarin (anticoagulant)
Glucosamine may enhance the effect of anticoagulants like warfarin, increasing the risk of bleeding. Close monitoring of INR is recommended.
Insulin and other antidiabetic agents
