Overview of the medicine
Relugolix 120 mg tablet is an oral gonadotropin-releasing hormone (GnRH) receptor antagonist used for the treatment of adult patients with advanced prostate cancer.
Uses & Indications
Dosage
Adults
Initial loading dose of 360 mg (three 120 mg tablets) on the first day, followed by 120 mg (one 120 mg tablet) orally once daily.
Elderly
No dosage adjustment is required for elderly patients.
Renal_impairment
Mild to moderate renal impairment: No dosage adjustment. Severe renal impairment (eGFR 15-29 mL/min): Initial loading dose of 180 mg on Day 1, followed by 60 mg orally once daily. End-stage renal disease (eGFR <15 mL/min) or on hemodialysis: Avoid use.
How to Take
Take orally once daily, with or without food, at approximately the same time each day. Swallow the tablet whole.
Mechanism of Action
Relugolix competitively binds to and blocks GnRH receptors in the anterior pituitary gland, which reduces the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). This leads to a rapid and sustained decrease in testosterone production by the testes.

Contraindications

Avoid concomitant use with strong P-gp inducers.
Storage
Store at room temperature (20°C to 25°C), with excursions permitted to 15°C to 30°C. Protect from moisture. Keep out of reach of children.
Overdose
No specific antidote for relugolix overdose. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Relugolix is contraindicated in pregnant women as it can cause fetal harm. Females of reproductive potential should use effective contraception during treatment and for 2 weeks after the last dose. It is unknown if relugolix is excreted in human milk, so breastfeeding is not recommended.
