Overview of the medicine
Rovator 10 mg Tablet contains Rosuvastatin, which belongs to a group of medicines called statins. It is used to lower high cholesterol levels and other fats (triglycerides) in the blood, reducing the risk of heart disease and stroke.
Uses & Indications
Dosage
Adults
Initial dose 5-10 mg orally once daily. Maximum 40 mg daily. Dosage should be individualized based on LDL-C levels and treatment goals.
Elderly
Initial dose 5 mg once daily for patients ≥70 years old.
Renal_impairment
For severe renal impairment (CrCl <30 mL/min), initial dose 5 mg once daily, maximum 10 mg once daily.
How to Take
May be taken with or without food at any time of day, but preferably at the same time each day.
Mechanism of Action
Rosuvastatin is a selective and competitive inhibitor of HMG-CoA reductase, an enzyme that catalyzes the rate-limiting step in cholesterol biosynthesis. It primarily acts in the liver to reduce cholesterol production and increase the number of LDL receptors on the surface of liver cells, enhancing the uptake and catabolism of LDL.
Pharmacokinetics
Onset
Lipid-lowering effects typically observed within 1 week, maximal effect usually by 2-4 weeks.
Excretion
Primarily excreted unchanged in feces (90%), with about 5% excreted in urine.
Half life
Plasma elimination half-life is approximately 19 hours.
Absorption
Well absorbed, absolute bioavailability about 20%. Peak plasma concentrations occur 3 to 5 hours after oral administration.
Metabolism
Limited hepatic metabolism (approximately 10%), primarily by CYP2C9 and CYP3A4 (to a lesser extent).
Side Effects
Contraindications
- •Hypersensitivity to rosuvastatin or any component of the formulation
- •Active liver disease, including unexplained persistent elevations of serum transaminases
- •Pregnancy and lactation
- •Concomitant use with cyclosporine
Drug Interactions
Warfarin
May enhance anticoagulant effect; monitor INR.
Gemfibrozil
Increases rosuvastatin exposure; avoid co-administration or limit rosuvastatin to 10 mg once daily.
Cyclosporine
Significant increase in rosuvastatin exposure (contraindicated).
Erythromycin
Decreases rosuvastatin exposure.
Antacids (aluminum and magnesium hydroxide)
Decreases rosuvastatin plasma concentration; administer antacids 2 hours after rosuvastatin.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
No specific antidote. Treatment should be symptomatic and supportive. Hemodialysis is not expected to significantly enhance rosuvastatin clearance.
Pregnancy & Lactation
Category X in pregnancy. Contraindicated during pregnancy and lactation due to potential for fetal harm and excretion into breast milk.
