Description
Overview of the medicine
Rupastar 10 mg Tablet contains Rupatadine, a second-generation antihistamine used to relieve symptoms of allergic rhinitis (e.g., sneezing, runny nose, itchy eyes) and chronic idiopathic urticaria (hives) in adults and adolescents 12 years and older.
Uses & Indications
Dosage
Adults
10 mg once daily, with or without food.
Elderly
No specific dose adjustment is generally required. Use with caution due to potential age-related decrease in renal or hepatic function.
Renal_impairment
Use with caution in patients with renal impairment. Not recommended in patients with severe renal impairment.
Hepatic_impairment
Use with caution in patients with hepatic impairment. Not recommended in patients with severe hepatic impairment.
How to Take
Take orally once daily, with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Rupatadine is a long-acting, selective peripheral H1-receptor antagonist. It also has antagonistic activity against platelet-activating factor (PAF), further contributing to its anti-allergic effects.
Pharmacokinetics
Onset
Onset of action typically occurs within 1 hour.
Excretion
Approximately 60.9% of the dose is excreted via feces and 34.6% via urine, mainly as metabolites.
Half life
The elimination half-life of rupatadine is approximately 5.9 hours (for the parent drug).
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations (Cmax) are reached approximately 0.75-1.5 hours post-dose. Extensive first-pass metabolism occurs.
Metabolism
Extensively metabolized by the cytochrome P450 system (primarily CYP3A4) into active and inactive metabolites, including desloratadine.
Side Effects
Contraindications
- •Hypersensitivity to rupatadine or any other component of the tablet

Drug Interactions
Statins
Rupatadine should be administered with caution when co-administered with statins as they are also metabolized by CYP3A4.
CNS depressants (e.g., alcohol, benzodiazepines)
May enhance the sedative effects of CNS depressants. Avoid concurrent use with alcohol.
Ketoconazole, Erythromycin, Clarithromycin, Itraconazole
Concurrent administration with strong CYP3A4 inhibitors may increase systemic exposure to rupatadine. Use with caution.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, tachycardia, and headache. In case of overdose, symptomatic and supportive treatment should be initiated. No specific antidote is available.
Pregnancy & Lactation
Pregnancy Category B. Use during pregnancy only if clearly needed after careful assessment of benefits and risks. It is not known whether rupatadine is excreted in human milk; use with caution in breastfeeding mothers.
