Overview of the medicine
Sasolin-D 0.4 mg Capsule is a combination medicine containing Tamsulosin and Dutasteride. It is used to treat symptoms of benign prostatic hyperplasia (BPH) such as difficulty in passing urine, frequent urination or needing to get up during the night to urinate. Tamsulosin helps relax the muscles in the prostate and bladder neck, while Dutasteride helps shrink the prostate gland.

Dosage
Adults
One capsule (Tamsulosin 0.4 mg / Dutasteride 0.5 mg) taken orally once daily, approximately 30 minutes after the same meal each day.
Elderly
No specific dosage adjustment is required for elderly patients.
Renal_impairment
No dosage adjustment is required for patients with renal impairment. Caution advised in severe impairment.
How to Take
Take orally, once daily, approximately 30 minutes after the same meal each day. Swallow the capsule whole with water; do not chew or crush it.
Mechanism of Action
Tamsulosin is an alpha-1A adrenergic receptor antagonist that selectively blocks receptors in the prostate, bladder neck, and prostatic urethra, leading to relaxation of smooth muscles and improved urine flow. Dutasteride is a 5-alpha reductase inhibitor that blocks the conversion of testosterone to dihydrotestosterone (DHT), which is responsible for prostatic growth. This leads to a reduction in prostate volume.

Tamsulosin: Symptomatic relief within days. Dutasteride: Prostate size reduction seen after 3-6 months.
Excretion
Tamsulosin: Primarily in urine as metabolites. Dutasteride: Primarily in feces as metabolites.
Half life
Tamsulosin: Approximately 9-15 hours. Dutasteride: Approximately 3-5 weeks.
Absorption
Tamsulosin is well absorbed, about 90% in fasting state. Dutasteride is well absorbed, bioavailability is approximately 60% with food.
Metabolism
Both primarily metabolized in the liver, Tamsulosin by CYP3A4 and CYP2D6, Dutasteride by CYP3A4 and CYP3A5.
Side Effects
Contraindications
- •Hypersensitivity to Tamsulosin, Dutasteride, other 5-alpha reductase inhibitors or any excipients.
- •Women, children, and adolescents.
- •Severe hepatic impairment.
Drug Interactions
Warfarin
Caution advised as Tamsulosin may displace warfarin from plasma protein binding sites.
Other Alpha-1 adrenergic blockers
Potential for enhanced hypotensive effects.
Strong CYP3A4 inhibitors (e.g., Ketoconazole)
May increase plasma concentrations of Tamsulosin and Dutasteride.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Overdose of Tamsulosin can lead to severe hypotensive effects. Dutasteride overdose symptoms are less defined. Treatment should be supportive and symptomatic. Gastric lavage and activated charcoal may be considered.
Pregnancy & Lactation
Sasolin-D is contraindicated in women, especially during pregnancy or lactation, due to the risk of dutasteride causing abnormalities in a male fetus. Women of childbearing potential should avoid contact with leaking capsules.
