Sedno 5 mg Tablet contains Zopiclone, a non-benzodiazepine hypnotic used for the short-term treatment of insomnia.
Uses & Indications
Dosage
Adults
The usual adult dose is 7.5 mg orally at bedtime. For Sedno 5 mg, it may be prescribed as an initial dose or for sensitive patients.
Elderly
Start with a lower dose of 3.75 mg; maximum 5 mg, taken orally at bedtime.

Start with a lower dose of 3.75 mg orally at bedtime. Dosage should be adjusted based on clinical response and tolerability.
How to Take
Take orally, immediately before bedtime. Do not take with food if faster onset is desired, although food does not significantly alter total absorption.
Mechanism of Action
Zopiclone potentiates the effects of GABA by binding to GABA-A receptors, leading to an increase in chloride ion influx and subsequent neuronal hyperpolarization, which enhances inhibitory neurotransmission in the central nervous system, thus promoting sleep.

Onset
Approximately 30 minutes after oral administration.
Excretion
Mainly excreted in urine (about 80%) and feces (about 16%), predominantly as metabolites.
Half life
The elimination half-life is 3.5 to 6.5 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract; peak plasma concentrations reached within 1 to 2 hours. Bioavailability is approximately 80%.
Metabolism
Extensively metabolized in the liver, primarily by CYP3A4 and CYP2C8, to two main metabolites: an active N-desmethylzopiclone and an inactive zopiclone-N-oxide.

Drug Interactions

May increase zopiclone plasma levels, potentially leading to increased sedative effects.
CYP3A4 inducers (e.g., rifampicin, carbamazepine, phenobarbital)

Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, lethargy, ataxia, hypotonia, hypotension, and respiratory depression. Treatment is supportive and symptomatic; gastric lavage may be considered if ingestion was recent. Flumazenil may be used in severe cases but is not routinely recommended for non-benzodiazepines.
Pregnancy & Lactation
Not recommended during pregnancy, especially in the third trimester due to risk of neonatal withdrawal symptoms. Zopiclone is excreted in breast milk; therefore, use during lactation should be avoided.
