Overview of the medicine
Selotin 50 mg Tablet contains Sertraline, a selective serotonin reuptake inhibitor (SSRI) used to treat various mental health conditions like depression, panic disorder, obsessive-compulsive disorder (OCD), and post-traumatic stress disorder (PTSD). It helps restore the balance of a natural substance (serotonin) in the brain.
Uses & Indications
Dosage
Adults
Initial dose is typically 50 mg once daily. May be increased up to 200 mg daily based on response and tolerability. For OCD, initial dose 50 mg daily, for panic disorder/PTSD/social anxiety, initial 25 mg daily then increased to 50 mg daily after one week.
Elderly
Lower initial doses and slower titration may be considered due to potential for increased sensitivity.
Renal_impairment
No dose adjustment is generally needed for mild to moderate renal impairment. Use with caution in severe renal impairment.
How to Take
Take orally, once daily, with or without food. It is recommended to take it at the same time each day.
Mechanism of Action
Sertraline selectively inhibits the reuptake of serotonin (5-HT) into presynaptic neurons, thereby increasing the concentration of serotonin in the synaptic cleft. This leads to enhanced serotonergic neurotransmission in the central nervous system.

Therapeutic effects may take 1-4 weeks to become apparent.
Excretion
Excreted in urine and feces, primarily as metabolites.
Half life
Approximately 26 hours (22-36 hours) for sertraline and 62-104 hours for its active metabolite, desmethylsertraline.
Absorption
Well absorbed after oral administration, peak plasma concentrations reached within 4.5 to 8.4 hours.
Metabolism
Extensive first-pass metabolism in the liver by multiple cytochrome P450 enzymes (mainly CYP2D6, CYP2C19, CYP2B6, CYP2C9, CYP3A4).
Side Effects
Contraindications
- Concomitant use with MAOIs (Monoamine Oxidase Inhibitors) or within 14 days of discontinuing an MAOI (due to risk of serotonin syndrome)
- Concomitant use with pimozide
- Hypersensitivity to sertraline or any component of the formulation

Pregnancy Category C. Use only if potential benefit outweighs risk to the fetus. Neonates exposed to SSRIs late in the third trimester have developed complications. Excreted in breast milk, use with caution during lactation; monitor infant for side effects.
