Overview of the medicine
SG-Plus is a combination medication containing Sildenafil Citrate and Dapoxetine Hydrochloride. It is used to treat erectile dysfunction and premature ejaculation in adult men.
Uses & Indications
Dosage
Adults
Typically, one tablet (Sildenafil 50/100 mg + Dapoxetine 30/60 mg) taken orally approximately 1-3 hours before anticipated sexual activity. Maximum once daily.
Elderly
No specific dose adjustment for elderly patients, but caution is advised due to potential for increased sensitivity.
Renal_impairment
No dose adjustment for mild to moderate renal impairment. Not recommended for severe renal impairment.
How to Take
Oral, with or without food. Avoid high-fat meals as it may delay Sildenafil absorption. Take with a full glass of water. Do not crush or chew.
Mechanism of Action
Sildenafil works by inhibiting PDE5, increasing blood flow to the penis for erection. Dapoxetine is an SSRI that delays ejaculation by increasing serotonin levels in the synaptic cleft.

Sildenafil: 30-60 minutes for erection. Dapoxetine: within 1-3 hours for ejaculation delay.
Excretion
Sildenafil: Metabolites excreted mainly in feces (approx 80%) and to a lesser extent in urine (approx 13%). Dapoxetine: Excreted mainly in urine (metabolites).
Half life
Sildenafil: approx 3-5 hours. Dapoxetine: approx 1.5-2 hours (terminal half-life).
Absorption
Sildenafil is rapidly absorbed, Tmax 30-120 min. Dapoxetine is also rapidly absorbed, Tmax approx 1-2 hours.
Metabolism
Sildenafil: Primarily hepatic via CYP3A4 and CYP2C9. Dapoxetine: Primarily hepatic via CYP2D6 and CYP3A4.
Side Effects
Contraindications
- Concomitant use with nitrates or nitric oxide donors
- History of hypersensitivity to Sildenafil or Dapoxetine
- Severe cardiovascular conditions (e.g., unstable angina, recent MI/stroke)
- Severe hepatic impairment
- Concomitant use with strong CYP3A4 inhibitors (for Sildenafil) or strong CYP2D6/CYP3A4 inhibitors (for Dapoxetine)
- History of mania or severe depression
Drug Interactions
Alcohol
Increased risk of dizziness, syncope, and adverse neurological effects with Dapoxetine.
Nitrates
Potentiation of hypotensive effects, potentially life-threatening.
