Description
Overview of the medicine
Stafen-0025 Eye Drop is an ophthalmic corticosteroid suspension used to treat inflammatory conditions of the eye, such as allergic conjunctivitis, anterior uveitis, and post-operative inflammation.
Uses & Indications
Dosage
Adults
Instill 1 drop into the conjunctival sac of the affected eye(s) 2-4 times daily. During the initial 24 to 48 hours, the dosage may be increased to 1 drop every hour. Treatment should not exceed 14 days unless specifically indicated by a physician.
Elderly
No specific dosage adjustment is generally required for elderly patients.
Renal_impairment
Not applicable for topical ophthalmic preparations due to minimal systemic absorption.
How to Take
For ophthalmic use only. Shake well before use. Tilt head back, pull down the lower eyelid to form a pouch, instill the drop, and close the eye gently for 1-2 minutes. Avoid touching the dropper tip to any surface to prevent contamination.
Mechanism of Action
As a corticosteroid, it inhibits the inflammatory response to various agents and may delay or slow healing. It acts by inducing phospholipase A2 inhibitory proteins, which control the biosynthesis of potent mediators of inflammation such as prostaglandins and leukotrienes.
Pharmacokinetics
Onset
Within a few hours for noticeable anti-inflammatory effects.
Excretion
Excreted primarily via urine if significant systemic absorption occurs, but mostly local elimination through ocular turnover.
Half life
Not well-defined for topical ophthalmic application due to minimal systemic absorption; estimated ocular tissue half-life around 4-6 hours for some similar compounds.
Absorption
Minimal systemic absorption occurs after topical ophthalmic administration. Locally absorbed into ocular tissues.
Metabolism
Primarily metabolized in ocular tissues and liver to inactive metabolites.
Side Effects
Contraindications
- Acute untreated purulent bacterial, viral, or fungal infections of the eye
- Epithelial herpes simplex keratitis (dendritic keratitis)
- Vaccinia, varicella, and most other viral diseases of the cornea and conjunctiva
- Mycobacterial infection of the eye
- Fungal diseases of ocular structures
- Hypersensitivity to the active ingredient or any excipients
Drug Interactions
Mydriatics/Cycloplegics
Potential for additive increase in intraocular pressure if used concurrently.
Other topical ophthalmic medications
Wait at least 5 minutes between administering different eye drops to prevent washout.
Storage
Store upright at room temperature (below 30°C). Protect from light and moisture. Do not freeze. Keep out of reach of children. Discard unused portion 28 days after opening.
Overdose
Overdose with topical ophthalmic preparations is unlikely due to limited systemic absorption. If accidental ingestion occurs, seek medical attention.
Pregnancy & Lactation
Use during pregnancy and lactation only if clearly needed and the potential benefits outweigh the potential risks. Prolonged or repeated use during pregnancy may be associated with an increased risk of intrauterine growth retardation. Consult a physician.

Clinical Trials
Clinical trials established the efficacy and safety of Fluorometholone in treating ocular inflammatory conditions. Specific trials for 'stafen-0025-eye-drop' would demonstrate bioequivalence and comparable therapeutic outcomes to reference Fluorometholone products.
Lab Monitoring
- Regular eye examinations to monitor intraocular pressure (IOP) if therapy extends beyond 10 days.
- Slit lamp examination to monitor for cataract formation or corneal changes with prolonged use.
Doctor Notes
- Advise patients on proper instillation technique and the importance of compliance.
- Monitor intraocular pressure regularly, especially if therapy exceeds 10 days.
- Examine the patient frequently for signs of secondary infection or delayed wound healing.
- Avoid abrupt discontinuation after prolonged use to prevent rebound inflammation.
- Not for use in fungal, viral (especially herpes simplex), or mycobacterial infections without appropriate concurrent anti-infective therapy.
Patient Guidelines
- Do not touch the dropper tip to any surface to avoid contamination.
- Shake the bottle well before each use.
- Remove contact lenses before use and wait at least 15 minutes before reinserting.
- Do not use if the solution changes color or becomes cloudy.
- Complete the full course of treatment as prescribed by your doctor, even if symptoms improve.
- Do not discontinue abruptly without consulting your doctor, especially after prolonged use.
Missed Dose Advice
If a dose is missed, apply it as soon as remembered. If it is almost time for the next dose, skip the missed dose and continue with the regular schedule. Do not double the dose.

Lifestyle Advice
- Maintain good eye hygiene to prevent further infection or irritation.
- Avoid rubbing eyes excessively.
