Overview of the medicine
Tabis 25 mg tablet contains Tadalafil, a phosphodiesterase-5 (PDE5) inhibitor used to treat erectile dysfunction (ED) and the symptoms of benign prostatic hyperplasia (BPH). It works by increasing blood flow to the penis to help men get and keep an erection, and by relaxing muscles in the prostate and bladder for BPH relief.
Uses & Indications
Dosage
Adults
For ED: 25 mg as needed, approximately 30 minutes before sexual activity. For BPH: 5 mg once daily. (Note: 25mg is not a standard Tadalafil dose for BPH, typically for ED).
Elderly
No dosage adjustment needed for elderly based solely on age, but renal function should be assessed.
Renal_impairment
For ED: Mild to moderate impairment, 10 mg once daily. Severe impairment, 5 mg once daily. For BPH: Mild to moderate impairment, 5 mg once daily. Severe impairment, not recommended.
How to Take
Tabis tablets are taken orally, with or without food. For ED, take it prior to anticipated sexual activity. For BPH, take it once daily at approximately the same time each day.
Mechanism of Action
Tadalafil inhibits phosphodiesterase-5 (PDE5), an enzyme responsible for the degradation of cyclic guanosine monophosphate (cGMP) in the corpus cavernosum. By inhibiting PDE5, Tadalafil increases cGMP levels, leading to smooth muscle relaxation and increased blood flow into the penis, resulting in an erection. For BPH, it helps relax smooth muscle in the prostate and bladder.

Onset
Onset of action typically within 30-60 minutes.
Excretion
Excreted predominantly in the feces (approximately 61% of the dose) and to a lesser extent in the urine (approximately 36% of the dose).
Half life
Approximately 17.5 hours.
Absorption
Rapidly absorbed after oral administration, with mean maximal plasma concentrations (Cmax) reached between 0.5 to 6 hours. Absorption is not affected by food.
Metabolism
Primarily metabolized by the cytochrome P450 (CYP) 3A4 isoform to a catechol metabolite, which is then glucuronidated.
Side Effects
Contraindications
- Concomitant use of nitrates in any form (due to potential for severe hypotension)
- Concomitant use of riociguat (a guanylate cyclase stimulator)
- Hypersensitivity to Tadalafil or any component of the tablet
Drug Interactions
Alcohol
May increase the risk of hypotension.
Nitrates
Concomitant use is contraindicated due to severe hypotensive effects.
Alpha-blockers
May lead to symptomatic hypotension; use with caution, initiate at lowest dose.
Antihypertensives
May have additive hypotensive effects.
CYP3A4 Inhibitors (e.g., Ketoconazole, Ritonavir)
Increase Tadalafil exposure; dose adjustment may be necessary.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, standard supportive measures should be adopted as required. Renal dialysis is not expected to accelerate clearance.

Tabis is not indicated for use in women. There are no adequate and well-controlled studies of Tadalafil in pregnant or lactating women.
