Description
Overview of the medicine
Talopram 10 mg Tablet is a selective serotonin reuptake inhibitor (SSRI) used to treat depression and various anxiety disorders by restoring the balance of serotonin in the brain.
Uses & Indications
Dosage
Adults
Initial dose is usually 10 mg once daily, increased to a maximum of 20 mg daily after 1-2 weeks if needed. For certain indications, the maximum may be 30 mg.
Elderly
A lower initial dose, typically 5 mg once daily, is recommended. Maximum recommended dose usually 10 mg daily.
Renal_impairment
No dose adjustment is generally required for mild to moderate renal impairment. Use with caution in severe impairment.
How to Take
Take orally, once daily, with or without food. Swallow the tablet whole with water. Do not chew or crush.
Mechanism of Action
Talopram selectively inhibits the reuptake of serotonin (5-HT) into presynaptic neurons, thereby increasing the concentration of serotonin in the synaptic cleft. This leads to enhanced serotonergic neurotransmission in the central nervous system.
Pharmacokinetics
Onset
Therapeutic effects may take 2-4 weeks to become fully apparent, though some patients may experience initial benefits sooner.
Excretion
Primarily excreted via urine (approximately 10% as unchanged drug) and feces.
Half life
Approximately 27-35 hours, allowing for once-daily dosing.
Absorption
Well absorbed orally, peak plasma concentrations typically achieved within 2-4 hours. Bioavailability is high, around 80%.
Metabolism
Extensively metabolized in the liver, primarily by CYP3A4 and CYP2C19. Active metabolites are formed but contribute minimally to overall effect.
