Description
Overview of the medicine
Tamsulosin Hydrochloride is an alpha-1 blocker used to treat the symptoms of an enlarged prostate (benign prostatic hyperplasia or BPH). It helps to relax the muscles in the prostate and bladder neck, making it easier to urinate.
Uses & Indications
Dosage
Adults
0.4 mg once daily, administered approximately 30 minutes after the same meal each day. May be increased to 0.8 mg once daily after 2 to 4 weeks if necessary.
Elderly
No dosage adjustment required for elderly patients.
Renal_impairment
No dosage adjustment required for patients with mild to moderate renal impairment. Use with caution in severe renal impairment (CrCl < 10 mL/min) as it has not been studied.
How to Take
Take one capsule orally once daily, approximately 30 minutes after the same meal each day. Do not crush, chew, or open the capsule.
Mechanism of Action
Tamsulosin selectively blocks alpha-1A and alpha-1D adrenergic receptors in the prostate, prostatic capsule, bladder neck, and prostatic urethra. Blockage of these receptors leads to relaxation of the smooth muscle in these tissues, thereby improving urine flow and reducing symptoms of BPH.
Pharmacokinetics
Onset
Improvement in urinary flow may be seen within 4 to 8 hours, but symptomatic relief may take 1 to 2 weeks.
Excretion
Primarily excreted in the urine (76% as metabolites, 4-9% unchanged drug) and feces (21% as metabolites).
Half life
Approximately 9-15 hours in plasma.
Absorption
Well absorbed from the gastrointestinal tract, nearly 90% bioavailable under fasting conditions. Food reduces absorption by 30% but increases consistency.
Metabolism
Extensively metabolized by cytochrome P450 enzymes in the liver (primarily CYP3A4 and CYP2D6).

Warfarin
