Description
Overview of the medicine
Tetclin 250 mg Capsule contains Tetracycline, a broad-spectrum antibiotic used to treat a variety of bacterial infections.
Uses & Indications
Dosage
Adults
General dosage: 250-500 mg every 6 hours. For acne: 250-500 mg daily in single or divided doses.
Elderly
No specific dose adjustment for elderly patients, but monitor renal function carefully.
Renal_impairment
Reduce dose in patients with impaired renal function (e.g., creatinine clearance < 80 mL/min); consult product labeling for specific guidelines.
How to Take
Take orally with a full glass of water, preferably 1 hour before or 2 hours after meals. Avoid taking with milk, dairy products, or antacids, as these can significantly reduce absorption.
Mechanism of Action
Tetracycline inhibits bacterial protein synthesis by reversibly binding to the 30S ribosomal subunit, thereby preventing the attachment of aminoacyl-tRNA to the ribosomal acceptor site.
Pharmacokinetics
Onset
Onset of action is typically within hours; peak plasma concentrations occur 2-4 hours after oral administration.
Excretion
Excreted mainly unchanged in urine (30-60%) and feces (via bile).
Half life
Approximately 6-11 hours.
Absorption
Incomplete and variable oral absorption (60-80%), significantly reduced by food, dairy products, and antacids.
Metabolism
Limited hepatic metabolism; primarily undergoes enterohepatic recirculation.
Side Effects
Contraindications
- Hypersensitivity to tetracyclines
- Pregnancy and lactation
- Children under 8 years of age due to risk of permanent tooth discoloration and inhibition of bone growth
- Severe renal impairment (unless dosage is appropriately adjusted)
Drug Interactions
Penicillins
Tetracyclines are bacteriostatic and may interfere with the bactericidal action of penicillins. Avoid concomitant use.
Methoxyflurane
Concurrent use may result in fatal renal toxicity.
Oral contraceptives
May reduce the effectiveness of oral contraceptives, leading to breakthrough bleeding or unintended pregnancy.
Anticoagulants (e.g., Warfarin)
Tetracycline may potentiate the effects of anticoagulants, requiring dosage adjustment.
Antacids, Iron preparations, Calcium, Magnesium, Bismuth subsalicylate
Significantly reduce tetracycline absorption. Administer at least 2 hours before or after tetracycline.
Storage
Store in a cool, dry place (below 30°C), away from direct light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, and diarrhea. Management is supportive, including gastric lavage or induced emesis, and symptomatic treatment. Hemodialysis is not effective.
Pregnancy & Lactation
Pregnancy Category D. Contraindicated during pregnancy and lactation due to the risk of permanent tooth discoloration and inhibition of bone growth in the fetus/infant.
