Description
Overview of the medicine
Tezo 200 mg Tablet contains Ofloxacin, a broad-spectrum antibiotic used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
Typical dosage is 200 mg to 400 mg every 12 hours, depending on the infection. For uncomplicated urinary tract infection, 200 mg orally every 12 hours for 3-7 days. For severe infections, dosage may be increased and duration extended.
Elderly
No specific dose adjustment is needed unless renal function is impaired. Monitor renal function.
Renal_impairment
Dose adjustment required. For creatinine clearance 20-50 mL/min, administer standard initial dose, then half the dose every 24 hours. For creatinine clearance <20 mL/min, administer standard initial dose, then half the dose every 48 hours.
How to Take
Tezo 200 mg Tablet should be taken orally, preferably with a full glass of water. It can be taken with or without food. Do not chew or crush the tablet. Complete the full course of treatment as prescribed by the doctor.
Mechanism of Action
Ofloxacin inhibits bacterial DNA gyrase and topoisomerase IV, enzymes essential for bacterial DNA replication, transcription, repair, and recombination, leading to bacterial cell death.
Pharmacokinetics
Onset
Within 1-2 hours
Excretion
Primarily excreted unchanged in the urine (70-80% of the dose); a small amount is excreted in feces.
Half life
Approximately 4-7 hours
Absorption
Rapidly and well absorbed from the gastrointestinal tract; bioavailability is approximately 95%. Peak plasma concentrations are achieved within 1-2 hours.
Metabolism
Minimally metabolized in the liver; about 5% of the dose is metabolized.
Side Effects
Contraindications
- Hypersensitivity to Ofloxacin or other fluoroquinolones
- History of tendon disorders related to fluoroquinolone use
- Epilepsy or other seizure disorders
- Children and adolescents (due to risk of joint damage)
Drug Interactions
Warfarin
May enhance anticoagulant effect, increasing bleeding risk. Monitor INR.
Theophylline
May increase plasma concentrations of theophylline, leading to toxicity. Monitor theophylline levels.
Corticosteroids
Increased risk of tendon rupture when co-administered.
NSAIDs (Nonsteroidal Anti-inflammatory Drugs)
May increase risk of CNS stimulation and convulsive seizures.
Antacids (containing magnesium, aluminum) / Iron supplements / Zinc supplements
Can reduce absorption of Ofloxacin. Take Ofloxacin at least 2 hours before or 6 hours after these agents.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, abdominal pain, dizziness, confusion, seizures, and prolonged QT interval. Treatment is symptomatic and supportive. Gastric lavage and activated charcoal may be considered. Hemodialysis and peritoneal dialysis are not effective.
Pregnancy & Lactation
Pregnancy Category C. Ofloxacin should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is excreted in human milk; therefore, a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
