Overview of the medicine
Tizaro 75 mg Injection is an NSAID used for rapid relief from acute pain, inflammation, and fever. It is administered via intramuscular route.
Uses & Indications
Dosage
Adults
Usually 75 mg once daily by deep intramuscular injection. In severe cases, 75 mg twice daily (morning and evening) can be given, preferably at different sites. Maximum daily dose is 150 mg.
Elderly
No specific dose adjustment is usually required, but caution is advised due to increased risk of side effects.
Renal_impairment
Use with caution; dose reduction may be necessary in severe impairment. Not recommended for patients with advanced renal disease.
How to Take
Administer by deep intramuscular (IM) injection. For IV administration, it must be diluted and infused slowly under strict medical supervision.
Mechanism of Action
It inhibits cyclooxygenase (COX-1 and COX-2) enzymes, thereby reducing the synthesis of prostaglandins, which are mediators of pain, inflammation, and fever.

Onset
Within 15-30 minutes for pain relief.
Excretion
Mainly through urine (approx. 60%) and bile (approx. 35%).
Half life
Approximately 1.5-2 hours.
Absorption
Rapidly and completely absorbed after intramuscular administration, reaching peak plasma concentrations within 10-30 minutes.
Metabolism
Primarily hepatic, undergoing hydroxylation and glucuronidation.
Side Effects
Contraindications
- Known hypersensitivity to the active substance or any excipients, or to other NSAIDs (e.g., aspirin)
- Active or history of recurrent peptic ulcer/hemorrhage
- Severe renal or hepatic impairment
- Severe heart failure
- Third trimester of pregnancy
- Coronary Artery Bypass Graft (CABG) surgery
Drug Interactions
Corticosteroids
Increased risk of gastrointestinal ulceration or bleeding.
Lithium and Methotrexate
Increased plasma levels of lithium and methotrexate.
Diuretics and ACE inhibitors
Reduced antihypertensive effect and increased risk of renal impairment.
Warfarin and other anticoagulants
Increased risk of bleeding.

Store below 30°C. Protect from light. Do not freeze.
Overdose
Symptoms may include nausea, vomiting, epigastric pain, gastrointestinal hemorrhage, diarrhea, disorientation, excitation, coma, drowsiness, tinnitus, and syncope. Acute renal failure and liver damage are possible. Management is symptomatic and supportive.
Pregnancy & Lactation
Avoid during the third trimester of pregnancy due to risk of premature closure of the fetal ductus arteriosus. Use with caution during first and second trimesters only if potential benefits outweigh risks. Excreted in breast milk in small amounts; use with caution during lactation.
