Overview of the medicine
Tolperisone is a centrally acting muscle relaxant used for the symptomatic treatment of spasticity and muscle spasm.
Uses & Indications
Dosage
Adults
Typically 150-450 mg daily, divided into 2-3 doses. For Tolgrin 200 mg tablet, one tablet (200 mg) once or twice daily, depending on individual response and severity.
Elderly
No specific dosage adjustment is usually required, but caution is advised. Start with a lower dose if necessary.
Renal_impairment
Use with caution in moderate renal impairment. Avoid in severe renal impairment (CrCl < 30 mL/min).
How to Take
Take orally, preferably after meals with a glass of water, to reduce gastrointestinal discomfort.
Mechanism of Action
Tolperisone reduces muscle tone via an action on the central nervous system. It primarily acts by stabilizing neuronal membranes and inhibiting voltage-gated sodium and calcium channels, thereby decreasing the excitability of motor neurons and preventing muscle contraction.
