Tromark 10 mg Tablet contains Atorvastatin, a statin, used to lower high cholesterol and triglyceride levels in the blood. It helps reduce the risk of heart attack, stroke, and other cardiovascular problems.
Uses & Indications
Dosage
Adults
Initial dose 10-20 mg once daily. Max dose 80 mg once daily. Adjust dosage based on LDL-C levels and patient response. For Tromark 10 mg: 1 tablet daily, preferably at the same time each day.
Elderly
No dosage adjustment required based on age alone. Follow adult dosage.
Renal_impairment
No dosage adjustment required for renal impairment.
How to Take
Tromark 10 mg Tablet can be taken orally, with or without food, at any time of the day. It is recommended to take it at the same time each day to maintain consistent drug levels.
Mechanism of Action
Atorvastatin selectively inhibits HMG-CoA reductase, an enzyme that catalyzes the early rate-limiting step in cholesterol biosynthesis. This inhibition reduces cholesterol production in the liver and increases the number of LDL receptors on liver cells, leading to increased clearance of LDL cholesterol from the blood.
Pharmacokinetics
Onset
Lipid-lowering effects are evident within 2 weeks, with maximum effect typically seen within 4 weeks.
Excretion
Primarily excreted in the bile following hepatic and/or extrahepatic metabolism. Less than 2% of the dose is recovered in the urine.
Half life
Approximately 14 hours for the parent drug; the active metabolites contribute to an extended plasma half-life of 20-30 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations usually occur within 1-2 hours. Absolute bioavailability is approximately 12%.
Metabolism
Extensively metabolized by cytochrome P450 3A4 (CYP3A4) to active ortho- and parahydroxylated metabolites and inactive beta-oxidation products.
Side Effects
Contraindications
- Hypersensitivity to Atorvastatin or any component of the tablet
- Active liver disease or unexplained persistent elevations of serum transaminases
- Pregnancy and lactation (or women of childbearing potential not using adequate contraception)
Drug Interactions
Warfarin
Potentiation of anticoagulant effect.
Erythromycin, Clarithromycin, Azithromycin, Itraconazole, Ketoconazole
Increased plasma concentrations of atorvastatin.
Cyclosporine, Gemfibrozil, Niacin, Protease inhibitors (e.g., Ritonavir)
Increased risk of myopathy/rhabdomyolysis.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
No specific antidote. In case of overdose, supportive measures should be taken as clinically indicated. Hemodialysis is not expected to significantly enhance atorvastatin clearance.
Pregnancy & Lactation
Atorvastatin is contraindicated during pregnancy and breastfeeding. It can cause fetal harm. Women of childbearing potential should use adequate contraception.
