Description
Overview of the medicine
Uniprost is an ophthalmic solution containing Latanoprost, used to reduce elevated intraocular pressure in patients with open-angle glaucoma and ocular hypertension.
Uses & Indications
Dosage
Adults
One drop in the affected eye(s) once daily in the evening.
Elderly
No special dosage adjustment is required for elderly patients.
Renal_impairment
No dosage adjustment is required for patients with renal impairment.
How to Take
Apply one drop into the conjunctival sac of the affected eye(s) once daily, preferably in the evening. If more than one topical ophthalmic drug is being used, they should be administered at least 5 minutes apart.
Mechanism of Action
Latanoprost is a prostaglandin F2α analogue that selectively increases the outflow of aqueous humor primarily through the uveoscleral pathway, thereby reducing intraocular pressure.
Pharmacokinetics
Onset
Onset of IOP reduction is about 3-4 hours.
Excretion
The metabolites are primarily excreted via the kidneys in urine.
Half life
The half-life of the latanoprost acid in the aqueous humor is approximately 17 minutes.
Absorption
Latanoprost is absorbed through the cornea where it is hydrolyzed to the biologically active acid form. Peak concentration in aqueous humor is reached about 2 hours after topical administration.
Metabolism
The active acid is metabolized primarily by fatty acid β-oxidation in the liver.
Side Effects
Contraindications
- Hypersensitivity to latanoprost or any other component of the formulation.
- Pregnancy (unless clearly necessary).
Drug Interactions
Other prostaglandin analogues
Concomitant use of two or more prostaglandin analogues is not recommended due to potential paradoxical increases in IOP.
Thimerosal-containing eye drops
Precipitation may occur when eye drops containing thimerosal are used in conjunction with latanoprost. If such drugs are used, they should be administered at least 5 minutes apart.
Storage
Store unopened bottles in a refrigerator (2-8°C). Once opened, store at room temperature (up to 25°C) for up to 4 weeks. Protect from light.
Overdose
Symptoms of latanoprost overdose may include ocular irritation and conjunctival hyperemia. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Pregnancy: Use only if the potential benefit justifies the potential risk to the fetus. Lactation: Latanoprost and its metabolites may be excreted in human milk. Caution should be exercised when administered to a nursing woman.
